Synthesis and Biological Evaluation of 1,2,4-Triazole and 1,3,4-Thiadiazole Derivatives as Potential Cytotoxic Agents

Abstract: A series of new 3-amino-5-sulfanyl-1,2,4-triazole and 2-amino-5-sulfanyl-1,3,4-thiadiazole derivatives have been synthesized and their cytotoxicities were evaluated on a panel of human cancer cell lines (BxPC-3, H1975, SKOV-3, A875, HCT116, etc.). The best one (compound 5m) exhibited activities with IC50 values ranging from 0.04 to 23.6 µM against nine human cancer cell lines. Further biological evaluation indicated that DNA replication was blocked by treatment with compound 5m in HCT116 cells.

“…Molecules 2020, 25, x 2 of 42 respectively. The compounds showed inhibition of growth of several tumor cell lines (ex vivo), with GI50 (the concentration causing 50% cell growth inhibition) values in the range of 0.74-10.0 μg/mL against the human colon cancer cell line HCT116, the human lung cancer cell line H460 and the human breast cancer cell line MCF-7.…”

“…Chemotherapy, radiation, and surgery are the major techniques among many, contemporarily applied for cancer treatment. Chemotherapy, the most used method (Shi et al, 2013 ), is regularly chosen in cancer cases where it has metastasised in the organism. The discovery of innovative anticancer drugs, presenting selectivity, and diminished side effects for regular cells, is a challenge (Ajmal et al, 2019 ).…”

“…On account of their unique structures and reactivities, 3-amino-2H-1,2,4-triazoles are widely utilised as intermediates to prepare various biologically important compounds, such as hypoglycemic agents, 1 [1,2,4]triazolo [1,5-a]pyrimidine bioactive derivatives, 2 tumour inhibitors [3][4][5][6] and selective inhibitors of TYK2 and JAK1 over JAK2 and JAK3. 7 1,2,4-Triazoles have an electron-rich bioisosteric ring that can mimic bioactive heterocyclic nuclei, such as imidazole, 8 pyrazole 9 and thiazole.…”
