Comprehensive Analysis for Histone Acetylation of Human Colon Cancer Cells Treated with a novel HDAC Inhibitor

Abstract: Extensive evidence suggests that dysregulation of histone lysine acetylation is intimately linked with the development of cancer in epigenetic level. Histone acetylation on lysine is regulated mainly by the "pencil"--Histone acetyltransferases (HATs) and the "eraser"--Histone deacetylases HDACs. Dramatic elevation of global histone deacetylation is considered as a biomarker for cancer. Therefore, current antitumor drug design often targets HDACs, inhibiting overexpressed HDAC in tumor cells with natural or syn… Show more

“…Epigenetic instability manifested as aberrant DNA methylation is observed in essentially all CRCs . Likewise, dysregulation of lysine acetylation in histones is intimately linked with the development of CRC and its metastasis . According to the epigenetic progenitor model of cancer epigenetic alterations affecting tissue specific differentiation are the prominent mechanism by which epigenetic changes cause cancer .…”

“…A proteomic study showed that the inhibition of Hsp90 in bladder cancer significantly affected the propionylation and butyrylation of core histones . Largazole‐7, a novel largazole derivative, was reported to alter the abundance of lysine methylation and butyrylation in histones, suggesting that targeting histone PTMs is a potential strategy for cancer therapy.…”

“…These can happen in different amino acid residues at several positions, although they are most common at histone N-terminal tails [ 29 , 30 ]. Mounting evidence suggests that aberrant histone lysine acetylation is a critical epigenetic driver of malignant transformation [ 31 , 32 ]. Histone lysine acetylation is regulated mostly by Histone acetyltransferases (HATs) and Histone deacetylases (HDACs).…”
