Enhanced beta-catenin expression and inflammation are associated with human ectopic tubal pregnancy

Abstract: A better understanding of the underlying mechanisms leading to ectopic pregnancies may contribute to our knowledge of the pathogenesis of tubal disorders and infertility and to the prevention of tubal ectopic pregnancy.

“…We also determined that expression of WNT/ CTNNB1 signaling‐related genes in the oviduct was regulated by E 2 . Recent findings indicate that the expression of CTNNB1 is increased in the ampulla region of fallopian tubes of patients with diseases, such as salpingitis, and ectopic pregnancy (41). Together with our findings, these observations suggest that aberrant expression of CTNNB1 in the fallopian tubes could result in ectopic pregnancy.…”

“…Mainly localized to cell-cell junctions, β-catenin plays an important role in intercellular adhesion, cell polarity, and architecture by binding to E-cadherin [21, 22]. Compared with non-pregnant tubal tissues, immunohistochemistry demonstrated that β-catenin was strongly expressed at tubal implantation sites in EPs following IVF-ET [23]. It is possible that the downregulation of CXXC5 increases expression of β-catenin, leading to a higher risk of EP in DOR.…”

“…Previous studies by us and others have shown that SLIT2‐ROBO1 signalling is involved in placental angiogenesis . VEGFA is the most potent angiogenic factor and is regulated directly by HIF1α . R5 treatment inhibits HIF1α and VEGFA expression.…”

“…Antibodies were used against the following targets: CK‐7 (Maixin Group, ShenZhen, China), E‐cadherin (Becton Dickinson, San Jose, CA, USA), SLIT2 (Santa Cruz Inc., Santa Cruz, CA, USA), Robo1 (Abcam, Cambridge, MA, USA), AP‐2α (Santa Cruz) and phospho‐histone H3 (PH3) (Santa Cruz). Immunohistochemistry and immunofluorescent staining and semiquantitative analysis by immunohistochemical reactive scores (IRSs) were conducted as described previously . The immunofluorescent stainings were counted manually at 4′,6‐diamidino‐2‐phenylindole (DAPI) and PH3 immunostaining merged images.…”
