Proximity to PML Nuclear Bodies Regulates HIV-1 Latency in CD4+ T Cells

Abstract: Nuclear bodies (NBs), characterized by the presence of the promyelocytic leukemia (PML) protein, are important components of the nuclear architecture, contributing to genetic and epigenetic control of gene expression. In investigating the mechanisms mediating HIV-1 latency, we determined that silenced but transcriptionally competent HIV-1 proviruses reside in close proximity to PML NBs and that this association inhibits HIV-1 gene expression. PML binds to the latent HIV-1 promoter, which coincides with transcr… Show more

“…Under this model, inflammatory stimuli increase the activity of the transcriptional activators NF-κB and NFAT, which are required for the transcription of viral genes (109, 110) and p53, which coincidently mediates apoptosis (111). Antioxidants may therefore silence transcriptional activity (109,110), while pro-oxidants, such as arsenic trioxide, may reactivate the viral reservoir (112). By the same token, both NO donors and endogenously generated NO may stimulate HIV reactivation (113).…”

“…Nevertheless, both studies indicate an unconventional mobility of PML-NBs, which appear to sense viral nucleoprotein complexes or replication intermediates, resulting in movement of the respective proteins even to the cytoplasm. Additionally, PML and hDaxx were implicated as contributing to the control of retroviral latency via epigenetic repression (12,13).…”
