Synergistic Interaction of Paclitaxel and Curcumin with Cyclodextrin Polymer Complexation in Human Cancer Cells

Abstract: The use of cytotoxic chemotherapic agents is the most common method for the treatment of metastatic cancers. Poor water solubility and low efficiency of chemotherapic agents are among the major hurdles of effective chemotherapy treatments. Curcumin and paclitaxel are well-known chemotherapic agents with poor water solubility and undesired side effects. In this study, a novel drug nanocarrier system was formulated by encapsulating curcumin and paclitaxel in poly(β-cyclodextrin triazine) (PCDT) for the therapy o… Show more

“…It was reported that more than 80% of patients diagnosed with lung cancer is suffering from metastatic diseases2. Although chemotherapy may suppress and prevent the spread of cancer cells, its acute toxicity coupled with poor solubility, adverse side-effects3 as well as drug resistance in tumors4 dispute its usage and remain a concern in the medical field. Combination therapy with the usage of a natural and pharmacologically safe anticancer drug together with a highly potent yet toxic commercial-anticancer agent is an attractive approach to address these limitations.…”

“…This yellow colored naturally occurring polyphenolic phytoconstituent is purified from the rhizome of the plant, Curcuma longa . Cur, despite being well-known for its anti-inflammatory, antioxidant and antibacterial properties, it is also an anticancer agent which was reported to be able to suppress and treat various types of malignancies35. Taxol or Paclitaxel (Ptx), on the other hand is a highly potent anticancer drug that is commercially available.…”

“…Because of their hydrophobic internal cavity and hydrophilic external surface, their unique molecular structure can form supramolecular host–guest complexes with various hydrophobic molecules [13], [14]. Based on their distinct physical and chemical properties [15], CDs are used in various fields, including in biological medicine [16]–[19], foodstuffs [20], [21], and environmental protection [22][24]. The polymer β-cyclodextrin (CDP) was synthesized by the reaction of β-CD with a cross-linking agent, epichlorohydrin [25].…”
