The S100 proteins have been extensively used as cancer biomarkers. The objectives of the present work were to immobilize the antibody anti-protein S100 to a net of semi-interpenetrated of polysiloxane and polyvinyl alcohol (POS-PVA discs), to investigate its capacity to capture S100 protein from serum and to quantify it by ELISA in sera from patients with prostatic adenocarcinoma (n = 15) and healthy individuals (n = 10). Also these values were compared to the S100 protein expression in the prostatic tissue through immunohistochemistry. The POS-PVA discs fixed about 92.8% of the offered antibody (7.75 microg of antibody per disc). The best values of the immobilized no-marked antibody anti-S100 and serum dilution were found to be 10 microg and 1:400, respectively. Optical density (OD) values for the sera of patients (0.425 +/- 0.042) with prostatic adenocarcinoma were significantly lower (P < 0.05) compared to those established for the healthy individuals (1.034 +/- 0.124). In the immunohistochemistry study no significant variations were observed in the number of positive S100 cells between prostatic adenocarcinoma (153.45 +/- 16.82) and normal prostate (147.04 +/- 18.98). These results showed a clear difference between S100 proteins expressed in tissue and presented in serum during the prostatic tissue neoplasic transformation. Sera analysis was more sensitive than immunohistochemistry S100 protein detection in the prostate tissue besides the advantage to be less invasive method.

Objectives: Pigmented cells, that contain inert, submicron-sized dietary particles, are a consistent feature of the base of human Peyer's patches (PP). We aimed (i) to phenotype these intestinal pigment cells (PC) in archival tissue specimens and (ii) to establish whether PC phenotype is altered in inflammatory conditions, especially Crohn's disease (CD). Methods: PCs contained within PP were identified by routine haematoxylin and eosin (H&E) staining and dark field microscopy of archival ileal sections for: adenocarcinoma (n = 16), colonic CD (n = 23), non-CD colitis (n = 10). Paraffin-embedded serial sections were graded for microscopic inflammation and then investigated immunohistochemically with antibodies against CD68, MAC387, CD14, CD11b, CD15, CD1a, S100, HLA-DR, CD86 and Cathepsin D. Analyses were by light and confocal microscopies. Results: The majority of PCs were CD68 positive (circa 80 %) with a minority (circa 20 %) staining for MAC387. Microparticles were mainly identified within cathepsin D negative lysosomal compartments. Histological inflammatory grade and disease type had no influence on cell phenotype. Conclusions: The microparticle-containing PCs of the PP base are mainly mature macrophages (CD68) of low metabolic and immunological activity. There is no evidence of differential PC phenotype or activation in differing disease states, including CD.

Middle ear adenomas (MEAs) are benign neoplasms along a spectrum with neuroendocrine neoplasms (carcinoid tumors). Immunohistochemical (IHC) staining for myoepithelial markers has not been reported in these tumors. The archives of the Cleveland Clinic, University of Virginia and Armed Forces Institute of Pathology were retrospectively searched for tumors arising within the middle ear with material available for IHC staining. Twelve cases of MEAs, four cases of jugulotympanic paragangliomas (JPGs), 10 cases of ceruminous adenomas (CAs) and four cases of ceruminous adenocarcinomas (CACs) were obtained. IHC staining was performed for smooth muscle actin (SMA), p63, S-100 protein, cytokeratin 5/6 (CK5/6), and cytokeratin 7 (CK7). The MEAs were positive for: CK7 (92 %, luminal), CK5/6 (92 %, abluminal), p63 (83 %, abluminal), and negative for SMA and S-100 protein. The JPGs were negative for CK7, CK5/6, p63 and SMA; S-100 protein highlighted sustentacular cells. The CAs were positive for: CK7 (100 %, luminal), CK5/6 (100 %, abluminal), S-100 protein (80 %, abluminal), p63 (100 %, abluminal), and SMA (90 %, abluminal). CACs demonstrated two patterns, (1) adenoid cystic carcinoma-type: positive for CK7 (100 %, luminal), CK5/6, S-100 protein, p63, and SMA (all 100 %, abluminal); and (2) conventional-type: CK7 (50 % luminal), and no CK5/6, SMA, S-100 protein, or p63 expression. The IHC profile of MEAs suggests that these tumors harbor at least two cell populations, including luminal and basal cells. However, unlike ceruminous adenomas, MEAs lack true myoepithelial differentiation given the absence of S-100 protein and SMA staining in all cases.
