2-Pyrone natural products and mimetics: isolation, characterisation and biological activity

Abstract: The review summarises natural products containing the 2-pyrone moiety. An emphasis has been placed upon the biological activity associated with 2-pyrones, particularly with respect to potential therapeutic or anti-microbial agents. Where appropriate, non-natural 2-pyrone analogues are discussed, particularly those derived from natural product lead compounds.

“…8 They exhibit a wide range of biological activities, such as antifungal, cytotoxic, neurotoxic and phytotoxic properties, and treatment of Alzheimer's disease and high cholesterol is discussed. 8,9,10 Furthermore, 4-hydroxy-2-pyrones have become one of the most important classes of anti-human immunodeficiency virus (HIV) agents in recent years. 8 These non-peptide compounds seem to be promising candidates for the treatment of AIDS.…”

In comparison with peptides, small a-pyrones are interesting lead candidates for synthetic strategies because of their lack of chiral centers.…”

“…3656-A1, a derivative of a class of fungal secondary metabolites known as 2-pyrones (Tomikawa et al 2000). 2-pyrones have been previously reported to have a broad range of biological activities (Hilgeroth 2002; Marrison et al 2002; McGlacken and Fairlamb 2005). Recently, rasfonin was shown to induce the death of ras-mutated pancreatic tumour (Panc-1) cells (Xiao et al 2014).…”
