2-Aminopyridine—a label for bridging of oligosaccharides HPLC profiling and glycoarray printing

Abstract: 2-Aminopyridine derivatives of oligosaccharides (OS-AP) were printed onto microchips by two different ways. The first method is based on direct covalent insertion of OS-AP in polyacrylamide gel 3D chip. The second method is based on conversion of OS-AP into more reactive OS-aminoalditol followed by covalent printing onto NHS-activated glass slides. This approach extends the range of saccharides suitable for covalent printing due to availability of commercial OS-AP and easy high-performance liquid chromatograph… Show more

“…In the past decade, several glycan microarrays have been developed to investigate the binding between CBPs and glycans [1][2][3][4][5][6][7][8][9]. These arrays make use of printed glycan slides in combination with fluorescence detection, and differ mainly in the way the glycans are immobilized, as reviewed by Culf et al [10].…”

“…1). Previously, a series of fluorophores for enhancing UV detection such as 2-aminopyridine [20], p-aminobenzoic acid [21], 4-aminobenzonitrile [22], 8-aminonaphthalene-1,3,6-trisulfonic acid (ANTS) [23], and 3-(4-carboxybenzoyl)quinoline-2-carboxaldehyde (CBQCA) [24] were linked to oligosaccharides by reductive amination with a reaction yield varying from good to excellent. Here, we have modified the procedure of Williams et al [25] usually used for short or branched oligosaccharides to correspond to long-chain polymeric carbohydrates.…”

“…In a recent work, Shilova et al [58] proposed an integrated approach to labeling, separation, characterization, and immobilization of oligosaccharides (OS). The protocol begins with modification of a mixture of OS by a fluorescent bifunctional reagent.…”
