Abstract:A series of 2,3-dihydro-3-(1-pyrryl)spiro[benzofuran-2,4'-piperidine]s (IV) and 2,3-dihydro-3-(1-pyrryl)spiro[benzofuran-2,3'-pyrrolidine]s (V) was synthesized and evaluated for cardiovascular activity. The majority of the compounds displayed good antihypertensive activity in the spontaneous hypertensive rat model at 50 mg/kg po. The synthesis of a novel spiro[1-benzofuran-2,4'-piperidin]-3-one scaffold has been achieved in five steps with an overall yield of 47%. The versatility of the spiropiperidine scaffold in the context of library synthesis is exemplified by selective and sequential derivatisation of the amino and aryl bromide functional groups, including the development of multi-step telescope reaction matrices.

Benzene ring fused to a five membered ring containing N or O atom exhibits wide range of biological activities.Benzofuran is present in polymers, pharmaceuticals, and bioactive natural products.Benzofuran is significant as insecticides, herbicides, anti-inflammatory and anti-viral agents.Indoles are well known scaffold showing noteworthy activities like anti-tumor, anti-viral, anti-microbial and anti-oxidant. Indole derivatives are valuable moiety in different therapeutically active drug molecules. On account of the biological significance of benzofuran and indole scaffold, this review describes the recent advances in microwave assisted synthesis of benzofurans and indoles, and covers literature from 2012-2020.
