1α,25-Dihydroxyvitamin D3 modulates the murine antibody response to pneumococcal capsular polysaccharide serotype 3 through IL-12

Abstract: 1a, 25(OH) 2 D 3 ] is a steroid hormone that regulates calcium metabolism. Besides, 1a,25(OH) 2 D 3 also has pronounced immunomodulatory effects: it strongly inhibits dendritic cell (DC) maturation and impairs IL-12 production. We studied the effect of 1a,25(OH) 2 D 3 on the antibody response to pneumococcal capsular polysaccharide (caps-PS) serotype 3. 1a,25(OH) 2 D 3 inhibited the IgG2a antibody response to caps-PS serotype 3. Besides, 1a,25(OH) 2 D 3 also inhibited IL-12 production and maturation of DC. … Show more

“…Polysaccharides can increase the expression of MHC class II and the co-stimulatory molecules CD80 and CD86. Various polysaccharides can affect the morphological maturity of DCs, upregulate IL-12 and GM-CSF, downregulate phagocytosis and antigen uptake activities of DCs or promote DC differentiation (Jeurissen et al, 2005;Khayrullina et al, 2008). These…”

“…The relationship being uncovered by scientists is complex, yet the underlying theme seems to be one of modulating the adaptive immune response while promoting innate immunity 10 . In regards to adaptive immunity, vitamin D affects the proliferation and differentiation of B and T cells and modulates immunoglobulin production 10 with one study showing suppression of antibody response to pneumococcal antigens in mice 11 . While these effects may have important roles in autoimmunity, it is not yet clear whether they will have a beneficial role in infection or sepsis.…”
