19F nuclear magnetic resonance study of fluoropyrimidine metabolism in strains of Candida glabrata with specific defects in pyrimidine metabolism

Abstract: Flucytosine (5-FC)-resistant strains were isolated from the haploid opportunistic pathogen Candida glabrata by UV-induced mutation and fluoropyrimidine selection. These strains were characterized biochemically, and the metabolism of fluorinated pyrimidines was studied by '9F nuclear magnetic resonance spectroscopy. No evidence was obtained from these studies for degradative metabolism of the fluorinated derivatives. In the parental susceptible strain of C. glabrata, 5-fluorouracil but not 5-FC was detected wit… Show more

“…4,7 In Vitro Cell Viability/Cytotoxicity Studies Cell viability was determined using the MTS assay, for which the soluble tetrazolium salt MTS is used. Briefly, 100% pretransduced human glioma U87 cells and rat glioma cell line RG2 cells were plated and cultured in 100-mm plates.…”

“…The quantitative nature of in vivo 19 F-MRS measurements has been confirmed with HPLC (10,11). Building on previous MRS studies of 5-FC metabolism in perfused yeast cultures (12)(13)(14)(15)(16), conversion of 5-FC to 5-FU by antibody-yeast CD conjugates has been observed with MRS (17).…”

“…With this technique it was possible to detect the uptake of 5-FC, its intracellular conversion to 5-FU, a relatively rapid metabolism of 5-FU to fluoronucleotides, but a relatively slow export of 5-FU to the medium. In the past 19 F-NMR has been used extensively to monitor the pharmacokinetics and metabGene Therapy olism of 5-FU in cells, animals and patients, [35][36][37][38][39][40] to study the metabolism of 5-FC in yeasts and bacteria, 41 and more recently to evaluate other models of CD gene therapy. 14,42 …”
