18β-Glycyrrhetinic Acid Ameliorates Neuroinflammation Linked Depressive Behavior Instigated by Chronic Unpredictable Mild Stress via Triggering BDNF/TrkB Signaling Pathway in Rats

Abstract: Evidence shows that inflammatory responses may encompass the onset of severe depressive illness. Traditionally used licorice contains 18β-glycyrrhetinic acid (18βGA), which has been demonstrated to reduce inflammation and oxidative stress. This study investigates the antidepressant effects of 18βGA and the underlying mechanism in rats exposed to chronic unpredictable mild stress (CUMS). Wistar rats were exposed to CUMS for 36 consecutive days to establish depression. 18βGA (10, 20, and 50 mg/kg) or fluoxetine … Show more

“…However, GA reduced the frequency of immobility episodes and total immobility time, and GAPC increased immobility latency in the abovementioned tests, and promoted grooming in the SS test. A mild antidepressant activity of GA has been demonstrated in recent animal [64,65], as well as clinical, studies [66]. As neuroinflammation is considered a major target for GA as an antidepressant agent [65], it is possible that PC could augment its effects by enhancing neuronal differentiation under inflammatory stress conditions [67].…”

“…However, the linkage of bidirectional changes in the BDNF level in the SH-SY5Y cells to the regulation of anxiety- and pain-like responses remains unclear because either increases or decreases in the level of BDNF associated with psychological stress could be dependent on the areas in the brain. Psychological stress increased the BDNF level in the anterior cingulate cortex and insular cortex [ 39 ] and decreased it in the prefrontal cortex [ 67 ] and hippocampus [ 68 ].…”
