Abstract: The maize p1 locus coincides with a major QTL (quantitative trait locus) determining levels of maysin, a C-glycosyl flavone that deters feeding by corn ear-worm. The p1 gene is tightly linked with a second gene, p2, and both genes encode similar Myb-domain proteins. We show here that maize cell cultures transformed with either the p1 or p2 genes expressed under a constitutive promoter accumulate transcripts for flavonoid biosynthetic genes, and synthesize phenylpropanoids and C-glycosyl flavones related to may… Show more

“…p1 and p2 are the principle regulatory genes of the flavone pathway necessary for maysin synthesis. Although under distinct tissue-specific regulation p1 and p2 encode exchangeable products [42]. Based on our model, which is enhanced by the p2-t sequence from teosinte, the progenitor of modern maize, the ancestral p gene might have had a similar expression pattern as p2 .…”

“…The duplication of an ancestral p gene gave rise to the p2 and p1 genes that encode functionally interchangeable proteins [42]. However, both genes differ in their regulatory sequences and hence in their tissue-specific expression.…”

“…In our study QTL were detected in the B73o2/o2 x CML161 testcross population for reducing aflatoxin located at marker UMC2096, which is at the same location as the gene p2. Tlie p2 has been reported the same as pi, including as affecting maysin production, suggesting that this QTL for reducing aflatoxin could be related to maysin (Zhang et al, 2003).…”

“…Genomic DNA from the single copy P1-rr allele was used as a reference, and the P1-wr cluster from B73 as an example of the multi-copy P1 allele. The primers used are specific for P1 , and do not hybridize on the P2 gene, an ortholog of P1 that is not involved in phlobaphene pigmentation [24]. Figure  3A shows the different high altitude landraces, inbred line B73 and P1-rr kernels at the same magnification, presenting different color or colorless of the pericarp and the stalk corresponding with the color of the cobs from which genomic DNA was extracted to analyze the P1 copy number.…”
