14-3-3 Proteins Regulate Protein Kinase A Activity to Modulate Growth Cone Turning Responses

Abstract: Growth cones regulate the speed and direction of neuronal outgrowth during development and regeneration. How the growth cone spatially and temporally regulates signals from guidance cues is poorly understood. Through a proteomic analysis of purified growth cones we identified isoforms of the 14-3-3 family of adaptor proteins as major constituents of the growth cone. Disruption of 14-3-3 via the R18 antagonist or knockdown of individual 14-3-3 isoforms switches nerve growth factor-and myelin-associated glycopro… Show more

“…While 14-3-3 proteins are known to regulate neuronal migration and axonal outgrowth (13, 26, 29, 60), their involvement in synapse formation and spine maintenance has not been fully investigated. Our study provides the first in vivo evidence of 14-3-3’s role in dendritic spine regulation.…”

“…p120RasGAP Is Required for Netrin-1-dependent Attraction of Embryonic Cortical Neurons-To determine whether p120RasGAP regulates netrin-1-dependent chemoattraction, we evaluated the effect of p120RasGAP depletion on cortical growth cone turning in response to a netrin-1 gradient using a Dunn chamber turning assay (45,50,51). The growth cones of cortical neurons electroporated with control siRNA turned randomly with no particular preference for any direction when exposed to a control PBS gradient (Fig.…”

“…For example, it is possible that A-kinase anchoring proteins (Tasken and Aandahl, 2004) mediate spatial targeting of PKA to axonal tips, as has been demonstrated in growth cone guidance of embryonic neurons (Han et al, 2007). Also, 14-3-3 proteins can downregulate PKA activity by stabilizing the interaction between regulatory and catalytic subunits of PKA for growth cone guidance (Kent et al., 2010). Similarly, several protein kinases can control PKA activity via regulating the interaction between its regulatory and catalytic subunits (Braun et al, 1991; Chaturvedi et al, 2006; Houslay, 2006).…”
