12S rRNA mitochondrial gene as marker to trace Sicilian mono-species dairy products

Abstract: A B S T R A C TFor a rapid, specific and sensitive identification of cows', ewes' and goats' milk in mono-species Sicilian dairy products, species-specific duplex-PCR protocol was applied. DNA samples from blood and experimental cheeses of Sicilian autochthonous breeds were extracted to amplify the 12S rRNA (and part of 16S rRNA in case of Ovis aries) mitochondrial species-specific gene fragment. The use of species-specific primers for Bos taurus, Capra hircus and Ovis aries species, after electrophoresis on a… Show more

“…Bottero et al (2003) developed a triplex PCR for identification of cow, goat and sheep milk in cheese samples, the sensitivity of the method was 0.5% of bovine in caprine milk. Tortorici et al (2016) eveloped a duplex PCR for the amplification of cow, ewe and goat DNA sequences specific to 12SrRNA gene. Substitution of buffalo with bovine milk in Mozzarella cheese was tested by species identification of bovine specific mitochondrial cytochrome b gene and mitochondrial cytochrome oxidase subunit I gene.…”

“…As a result, easy, cheap, accurate, and simultaneous identification of the target species can be achieved with a good sensitivity threshold (0.5%) in milk and cheeses instead of IEF. Recently, Tortorici et al (2016) described a sensitive speciesspecific duplex-PCR method to detect cow, ewe, and goat dairy products simultaneously, amplifying 12S and 16S rRNA genes with 0.1% threshold. The speciesspecific primers were tested in Italian breeds for both studies, so possible nucleotide polymorphisms between the different goat breeds should be taken into account, as they could affect PCR amplification success.…”

“…Mitochondrial DNA (mtDNA) polymorphisms have been widely used to determine phylogenetic relationships in cattle breeds. The results of this 12S rRNA sequence have been used to identify the type of milk in the cattle of sheep and goats (Tortorici et al, 2016), to identify the types of livestock meat , to identify the presence of a mixture of dog and rat meat in beef (Cahyadi et al, 2019), and to detect pork and beef in food products (Roy et al, 2021). Based on these reasons, the 12S rRNA gene can be used as a marker of genetic diversity, and until now, there has been no report on Bali cattle in particular.…”
