Abstract

Melanoma is currently the most highest mortality rate of skin tumors, but the treatment of melanoma is limited.The global incidence of melanoma continues to increase every year. This tumor has highly aggressive and metastatic potential.Therefore, inhibition of tumor cell invasion may provide an effective therapy for melanoma. Here we report the Nicotinamide ,also named nicotinic acid (NA) ,which can inhibits melanoma cell invasion in vitro. Treatment of the B16-F10 cell with NA in vitro results in reduced invasion, and has concentration depends on the relationship,we found that inhibition of cell migration was most obvious when the concentration of niacin was 10 mM. At the molecular level, transcription of the adherens junction protein E-cadherin is increased, leading to accumulation of E-cadherin protein at the cell-cell boundary.The expression of transcription factor snail2 and twist which can promote tumor cell migration are significantly decreased.And we also found NA significantly inhibited the migration of b16-f10 cells, but did not affect its survival. These results suggest that NA treatment may be developed into a potential therapy for melanoma.
