11β-Hydroxysteroid Dehydrogenase Type 1 Expression Is Increased in the Aged Mouse Hippocampus and Parietal Cortex and Causes Memory Impairments

Abstract: Increased neuronal glucocorticoid exposure may underlie interindividual variation in cognitive function with aging in rodents and humans. 11␤-Hydroxysteroid dehydrogenase type 1 (11␤-HSD1) catalyzes the regeneration of active glucocorticoids within cells (in brain and other tissues), thus amplifying steroid action. We examined whether 11␤-HSD1 plays a role in the pathogenesis of cognitive deficits associated with aging in male C57BL/6J mice. We show that 11␤-HSD1 levels increase with age in CA3 hippocampus and… Show more

“…The high vitamin A status of LOU/C rats may confer protection against 11b-HSD1-mediated deleterious action on cognitive performance with age. Indeed, Holmes et al (2010) showed that an increased hippocampal expression of 11b-HSD1 was responsible for memory impairments in mice. Partial deficiency or short-term inhibition of 11b-HSD1 improves cognitive function in ageing mice (Sooy et al 2010).…”

“…The inhibitory effect of retinoids on 11b-hydroxysteroid dehydrogenase 1 (11b-HSD1) expression, an enzyme that regenerates active GCs within cells, has been reported in liver and visceral fat of WNIN/Ob obese rats (Sakamuri et al 2011), in adipose tissue of mice (Sakuta et al 2006) and in vitro in myotubes (Aubry & Odermatt 2009). This is interesting in regard to the fact that 11b-HSD1 increases in the aged hippocampus and its elevated expression correlates with impaired spatial memory (Holmes et al 2010).…”

“…Subsequently, a growing body of evidence has suggested that the age-related increase in 11β-HSD1 occurs in a variety of cells and tissues, including lymphocytes (28), bone (29), brain (30), and adipose (31), implicating a role for 11β-HSD1 in systemic aging. In these tissues, increased local GC activation is postulated to contribute to the increased risk of fracture and osteoporosis and impaired cognitive function associated with aging.…”

“…Recent studies have shown that HSD1 is not strictly limited to metabolic processes. Aged C57BL/6 mice show watermaze deficits that correlate with increased HSD1 expression in the hippocampus and forebrain; overexpression of HSD1 similarly impaired performance (Holmes et al, 2010). Conversely, aged HSD1 knock-out mice have improved cognition and increased long-term potentiation relative to age-matched controls, suggesting a neuroprotective effect of HSD1 inhibition (Yau et al, 2001(Yau et al, , 2007.…”
