111 Single Equine Embryo Lipid Fingerprinting by Mass Spectrometry

Abstract: Matrix-assisted laser desorption-ionization mass spectrometry (MALDI-MS) allows the lipid profile study of individual mammal embryos. The data collection is rapid, highly sensitive, can tolerate some level of impurities, and is easy to interpret. The aim of this study was to report the lipid profile obtained from a single equine embryo by MALDI-MS. Follicles ≥30 mm in diameter were monitored daily until ovulation (Day 0). The insemination was performed close to ovulation with fresh diluted semen, and the embry… Show more

“…MALDI-MS also allows single oocytes to be analysed, avoiding the pooling of tens to hundreds gametes for one single measurement. The use of MALDI-MS and related mass spectrometric protocols of mammalian oocytes and embryos is being rapidly incorporated in embryological research (Gonc¸alves et al 2011;Ferreira et al 2012a,b).…”

“…Of the 20 potentially relevant articles, one was an in vitro experimental study [ 46 ]; three articles were excluded because of the wrong population (cumulus cells [ 47 ] or older fetus [ 48 , 49 ]); and 10 published either no MSI results, only MS data [ 50 , 51 , 52 , 53 , 54 , 55 , 56 , 57 , 58 ], or only proteomics of the embryos [ 59 ]. One abstract was also excluded because no data extraction could be performed [ 60 ].…”

“…Although the topic is highly current, only five publications were found in the systematic literature search that met all our inclusion criteria. Several former studies investigated the role of lipids in oocytes [ 66 , 67 ], early stages of embryo development [ 52 , 56 , 58 , 66 , 68 , 69 , 70 , 71 ], placenta in late pregnancy [ 72 , 73 ], as well as endometrial tissues during the window of implantation [ 74 ], and in endometrial fluid [ 75 ]. However, in these studies, lipid composition was mostly investigated with MS and not with MSI.…”

“…(7) Lipid profiles of single oocytes and embryos were evaluated by mass measurement of phospholipid profiles using matrix-assisted laser desorption/ ionization mass spectrometry (MALDI-MS) fingerprinting. (8,9) The characteristics of a single bovine embryo after cryopreservation were also measured by MALDI-MS. (10) A single oocyte and single embryo lipid were also analyzed by a more direct and simpler MS technique of desorption electrospray ionization mass spectrometry (DESI-MS). (11) However, as these mass measurements are based on destructive analysis techniques, the analyzed embryos are not able to progress to transplantation.…”
