Interleukin 10 (IL10) and transforming growth factor β1 (TGFβ1) gene polymorphisms in persistent IgE-mediated cow's milk allergy

Abstract: OBJECTIVES:The aim of this cross-sectional study was to evaluate whether interleukin 10 (IL10) and transforming growth factor β1 (TGFβ1) gene polymorphisms were associated with persistent IgE-mediated cow's milk allergy in 50 Brazilian children. The diagnostic criteria were anaphylaxis triggered by cow's milk or a positive double-blind, placebo-controlled food challenge. Tolerance was defined as the absence of a clinical response to a double-blind, placebo-controlled food challenge or cow's milk exposure.METHO… Show more

“…The rs1800469 is located within a YY1 (Ying Yang 1) consensus binding site and is known to regulate transcriptional activity (Silverman et al, 2004). The rs1800469 SNP has been associated with elevated gene expression and circulating TGFB1 and IgE levels (Grainger et al, 1999; Jacob et al, 2013). Increased transcriptional activity of the T allele, as compared to the C allele, has been reported (Grainger et al, 1999).…”

“…The majority (n = 11) of included studies were conducted in only “Caucasian,” “European” or “White” populations, whereas 11 studies were carried out in predominantly Caucasian populations alongside other ethnicities (“Asians,” “Mixed” and “African American”) . Four others were carried out in Japanese populations, one was carried out in a Taiwanese population, and there were five studies where ethnicity was not mentioned …”

“…Chen et al and Jacob et al investigated variants at the gene encoding interleukin 10 ( IL10 ) in relation to any food and cow's milk allergy, respectively. A common SNP investigated by both studies is the ‐1082 A/G (rs1800896) variant, a 2 kilo base pair (kb) upstream variant.…”

“…A more recent study in Brazilian children examined several IL10 polymorphisms and a variant in the TGFβ1 gene. The only significant association found was that of IL-10 polymorphism-1082G/A with cow’s milk allergy [21]. …”
