Genome Mining of EndophyticStreptomyces wadayamensisReveals High Antibiotic Production Capability

Abstract: The actinobacteria Streptomyces wadayamensis A23, an endophitic strain, was recently sequenced and previous work showed qualitatively that the strain inhibits the growth of some pathogens. Herein we report the genome analysis of S. wadayamensis which reveals several antibiotic biosynthetic pathways. Using mass spectrometry, we were able to identify desferoxamines, several antimycins and candicidin, as predicted. Additionally, it was possible to confirm that the biosynthetic machinery of the strain when compare… Show more

“…XY006, an endophyte isolated from Tea ( Camellia sinensis ) had 27 possible secondary metabolite biosynthetic gene clusters encoding for antimycin, melanin, informatipeptin, albaflavenone, and roseoflavin (Vazquez-Hernandez et al, 2017 ). Genome mining of Streptomyces wadayamensis A23, an endophytic strain showed the presence of biosynthetic gene clusteres for antimycins and candicidin (Angolini et al, 2016 ).…”

“…Close fragmentation pattern was reported for desferrioxamine B complexed with Fe(III) (difference of 29 Da in the mass fragments due the difference of mass between Al and Fe) 53 . For compound 4, mass fragmentation yielded to fragments at m/z 423.2225 and 223.1101, this fragmentation pattern corresponds to the typical neutral losses of molecules of succinylcadaverine (200.1160 Da) 54 . Compounds and 4 were recovery using all types of extraction methods when GYE-ASW medium was used to bacterial growth.…”

“…Genome mining tools have made it more convenient to look for innovations in natural product discovery with majority of the bioactive compounds biosynthetic pathway of polyketides governed by a complex enzyme system, called polyketide synthase encoded by PKS gene cluster [121,122]. Available whole genome draft of endophytic actinobacteria also revealed the presence of PKS and NPRS genes suggesting that these microbes are the possible source for many novel bioactive compounds [123,124]. Screening for the presence of bioactive secondary metabolites in actinobacteria can be done using a high throughput method based on gene clusters.…”
