Box–Behnken Response Surface Design of Polysaccharide Extraction from Rhododendron arboreum and the Evaluation of Its Antioxidant Potential

In the present investigation, the ultrasound-assisted extraction (UAE) conditions and optimization of Rhododendron arboreum polysaccharide (RAP) yield were studied by a Box–Behnken response surface design and the evaluation of its antioxidant potential. Three parameters that affect the productivity of UAE, such as extraction temperature (50–90 °C), extraction time (10–30 min), and solid–liquid ratio (1–2 g/mL), were examined to optimize the yield of the polysaccharide percentage. The chromatographic analysis revealed that the composition of monosaccharides was found to be glucose, galactose, mannose, arabinose, and fucose. The data were fitted to polynomial response models, applying multiple regression analysis with a high coefficient of determination value (R2 = 0.999). The data exhibited that the extraction parameters have significant effects on the extraction yield of polysaccharide percentage. Derringer’s desirability prediction tool was attained under the optimal extraction conditions (extraction temperature 66.75 °C, extraction time 19.72 min, and liquid–solid ratio 1.66 mL/g) with a desirability value of 1 yielded the highest polysaccharide percentage (11.56%), which was confirmed through validation experiments. An average of 11.09 ± 1.65% of polysaccharide yield was obtained in optimized extraction conditions with a 95.43% validity. The in vitro antioxidant effect of polysaccharides of R. arboreum was studied. The results showed that the RAP extract exhibited a strong potential against free radical damage.

The purpose of this research is to optimize several variables, including fiber length, volumetric fiber percentage, and sodium hydroxide (NaOH) solution concentration, which influence the bending and compression behavior of cementitious composites reinforced with plant-based fibers from Agave americana. Samples of composites based on fibers extracted from Agave americana and a cement matrix were manufactured and prepared according to a reference mortar. Box-Behnken Design (BBD) of Response Surface Methodology (RSM) was employed to build an experimental design to explore their mechanical behavior and to construct a set of mathematical models predictive of their behavior. The first stage of the work includes the extraction and processing of Agave americana fibers and the preparation of cement and Agave americana mortar specimens. The second phase aims to establish mathematical models to forecast the workability of the cement mortar and its strength at 28 days while emphasizing the correlations and interactions between the different components through the Design Expert software.

O objetivo deste trabalho foi o aproveitamento integral do umari (Poraqueiba sericea Tulasne) para a elaboração de farinhas e incorporação em mistura para pão de frigideira sem glúten, verificando o impacto nos parâmetros de qualidade e compostos bioativos. Para isso foram analisados parâmetros físico-químicos e de qualidade (proteínas, lipídios, carboidratos, valor energético, beta caroteno, ácidos graxos, minerais). Também foi feita a vida de prateleira do produto ao longo de 180 dias (pH, cor, umidade, acidez, AW, índice de peróxidos e contagem de bolores, leveduras e infestação por insetos). As misturas enriquecidas com polpa e resíduo de umari liofilizado apresentaram teores lipídicos mais altos (10,89g e 9,07g respectivamente) e maiores valores energéticos (399,83kcal e 386,01kcal, respectivamente). As misturas enriquecidas apresentaram valores ideais de pH, cor, umidade, acidez, AW, índice de peróxidos, contagem de bolores, leveduras e infestação por insetos ao longo de 180 dias. A adição de polpa de uma liofilizada, resíduo de umari liofilizado, farinha de polpa de umari e farinha de resíduo de umari levou a um aumento de ácido oleico em relação ao pão controle. Também pudemos observar um aumento de magnésio nas misturas enriquecidas com os diversos pós e farinhas de umari e uma diminuição nos teores de sódio. A suplementação de pós e farinhas de umari na mistura de pão sem glúten foi uma abordagem útil e eficiente para obter um produto de qualidade e com bons parâmetros físico-químicos no armazenamento durante seis meses.
