PURPOSELimited information is available on multiple myeloma (MM), chronic lymphocytic leukemia (CLL), and non-Hodgkin lymphoma (NHL) management in Latin America. The primary objective of the Hemato-Oncology Latin America (HOLA) study was to describe patient characteristics and treatment patterns of Latin American patients with MM, CLL, and NHL.METHODSThis study was a multicenter, retrospective, medical chart review of patients with MM, CLL, and NHL in Latin America identified between January 1, 2006, and December 31, 2015. Included were adults with at least 1 year of follow-up (except in cases of death within 1 year of diagnosis) treated at 30 oncology hospitals (Argentina, 5; Brazil, 9; Chile, 1; Colombia, 5; Mexico, 6; Panama/Guatemala, 4).RESULTSOf 5,140 patients, 2,967 (57.7%) had NHL, 1,518 (29.5%) MM, and 655 (12.7%) CLL. Median follow-up was 2.2 years for MM, 3.0 years for CLL, and 2.2 years for NHL, and approximately 26% died during the study observation period. Most patients had at least one comorbidity at diagnosis. The most frequent induction regimen was thalidomide-based chemotherapy for MM and chlorambucil with or without prednisone for CLL. Most patients with NHL had diffuse large B-cell lymphoma (DLBCL; 49.1%) or follicular lymphoma (FL; 19.5%). The majority of patients with DLBCL or FL received rituximab plus cyclophosphamide, doxorubicin, vincristine, and prednisone.CONCLUSIONThe HOLA study generated an unprecedented level of high-quality, real-world evidence on characteristics and treatment patterns of patients with hematologic malignancies. Regional disparities in patient characteristics may reflect differences in ethnoracial identity and level of access to care. These data provide needed real-world evidence to understand the disease landscape in Latin America and may be used to inform clinical and health policy decision making.

BackgroundMultiple myeloma is a plasma cell neoplasm with acquired genetic abnormalities of clinical and prognostic importance. Multiple myeloma differs from other hematologic malignancies due to a high fraction of low proliferating malignant plasma cells and the paucity of plasma cells in bone marrow aspiration samples, making cytogenetic analysis a challenge. An abnormal karyotype is found in only one-third of patients with multiple myeloma and interphase fluorescence in situ hybridization is the most useful test for studying the chromosomal abnormalities present in almost 90% of cases. However, it is necessary to study the genetic abnormalities in plasma cells after their identification or selection by morphology, immunophenotyping or sorting. Other challenges are the selection of the most informative FISH panel and determining cut-off levels for FISH probes. This study reports the validation of interphase fluorescence in situ hybridization using CD138 positive cells, according to proposed guidelines published by the European Myeloma Network (EMN) in 2012.MethodBone marrow samples from patients with multiple myeloma were used to standardize a panel of five probes [1q amplification, 13q14 deletion, 17p deletion, t(4;14), and t(14;16)] in CD138+ cells purified by magnetic cell sorting.ResultsThis test was validated with a low turnaround time and good reproducibility. Five of six samples showed genetic abnormalities. Monosomy/deletion 13 plus t(4;14) were found in two cases.ConclusionThis technique together with magnetic cell sorting is effective and can be used in the routine laboratory practice. In addition, magnetic cell sorting provides a pure plasma cell population that allows other molecular and genomic studies.

Effect of transgenic cotton continuous cropping on soil bacterial community

Purpose In agricultural practices, continuous cultivation of genetically modified crops with high commercial value has a definite impact on soil microbial diversity. Soil microorganisms directly define the operational degree and function realization of the soil ecosystem. To understand the safety of environmental release, we studied the effects of continuous cropping of transgenic cotton on the diversity of bacterial communities in the rhizosphere soil. Methods We have applied a high-throughput sequencing method and compared the bacterial community structure as well as diversity of rhizosphere soil of the transgenic cotton line (25C-1) and its parent cotton line (TH2). Result Structural analysis of the bacterial community showed that Arthrobacter and Sphingomonas are significantly enriched after continuous cropping of transgenic cotton lines and had a positive impact on the soil’s ecological environment. Interestingly, parameters of the physical and chemical properties of soil used for the continuous cropping of the two cotton lines for 3 consecutive years show no detectable change, other than total nitrogen. Notably, Spearman’s correlation analysis suggests that total nitrogen is the key environmental factor that affects the bacterial community of the soil used to cultivate the transgenic cotton. Conclusion We did not find a notable difference in species diversity between the two samples. However, the proportions of beneficial bacteria (Arthrobacter and Sphingomonas) increased and the total nitrogen content has changed in 3 years. These results provide necessary insights into the function and role of bacteria in transgenic cotton. This study will help future investigators assess the potential ecological risks of genetically modified plants.
