Development and validation of stability indicating spectrophotometric and HPTLC methods for determination of acemetacin

Abstract: Three simple and sensitive methods were developed for the determination of acemetacin (ACM) in presence of its degradation product, indomethacin (DEG). Several methods were evaluated and are presented. Method A was based on measuring the peak amplitude of the first derivative of the ratio spectra 1 DD at 244 nm. In method B, mean centering of ratio spectra method (MCR) was applied, which depends on measuring the mean centered values of ratio spectra of ACM at 234 nm. Method C provided separation of ACM from it… Show more

“…The system suitability test (SST) is an essential part of the analytical method that is used to verify effectiveness of the final operating system [24]. The test was performed by injecting the standard sample in triplicate and the SST parameters were calculated as reported by USP [25], which include capacity factor (k'), selectivity factor (α), resolution factor (Rs), column efficiency (number of theoretical plates, N) [26].…”

“…Thin layer chromatography has become a well-established technique for the assay of drugs either in binary or in multicomponent mixtures [20][21][22][23][24][25][26][27]. The proposed method is based on the difference in the retardation factor (RF) between the CRM, PSH and MP.…”

“…The developed MCR method depends on the mean centering of the ratio spectra, it gets rid of the derivative steps and therefore signal-to-noise ratio is increased [29]. This method is able to resolve binary and ternary mixtures in complex samples with unknown matrices [30].…”
