The cytotoxic activity of Linum grandiflorum leaves

Abstract: The CHCl3 and MeOH fractions of the leaves of Linum grandiflorum, showed cytotoxic activity against EL4 (Murine Leukemia) cell line with IC50 = 60 and 250 g/mL respectively. Bioassay guided fractionation and isolation (BGFI) of the MeOH fraction resulted in the isolation of two new cytotoxic compounds 1 [luteolin 7-Oα-D-(6```-E-feruloyl)glucopyranosyl (1→2)-β-Dglucopyranoside] and 6 [2-[(3`-isopropoxy-O-β-D-glucopyranosyl)oxy]-2-methylbutane nitrile] against EL4 with IC50 = 0.2 and 0.3 M/mL respectively, tog… Show more

“…Leaves of L. grandiflorum were investigated previously by our group for its phytoconstituents, which resulted in the isolation of flavonoids, cyanogenic glycosides, and lignans (Mohammed et al, ), moreover, the cytotoxicity (Mohammed, Chen, Zhai, & Ibrahim, ) and anti‐HIV‐1 activity (Mohammed et al, ) of both leaves and seeds “MeOH and CHCl 3 extracts” were also evaluated and recorded promising results.…”

“…The benzylideneacetophenones (chalcones) (3a-d) were prepared in high yields by Claisen-Schmidt reaction of acetophenones (1a-d) with benzaldehyde (2) in the presence of sodium hydroxide in water-ethanol mixture [13,14]. The structures of the chalcones were confirmed by IR and 1 H NMR spectral studies.…”

“…Aryltetrahydronaphthalene-type lignans as podophyl lotoxin and its derivatives are important natural products in the armamentarium of antineoplastic agents. The biological assessment of podophyllotoxin was followed by discovery of its mode of action and culminated in the synthesis of the anticancer drugs etoposide and teniposide [2]. Hence, current research is focused on synthesizing novel compounds and to study their structure activity relationship [3].…”

“…62 When evaluated for cytotoxic activity, 114 had an IC 50 value of 0.3 μM against the EL 4 cell line, compared to the positive control, the cytotoxin thapsigargin, which had an IC 50 value of 1.9 μM. 63 The first report of a cyanogenic glycoside in mistletoes was that of 2-methylpropionitrile-2-O-(6-O-galloyl)-β-D-pyranoglucoside, named as linamarin gallate 115 (Figure 12), isolated from the leafy twigs of the Nigerian species Loranthus micranthus L. (Loranthaceae), parasitic on Hevea brasiliensis Müll.Arg. (Euphorbiaceae).…”
