Malaria is one of the major health problems in developing countries. The disease kills a large number of people every year and also affects financial status of many countries. Resistance of the plasmodium parasite, the causative agent, to the existing drugs, including chloroquine, mefloquine, and artemisinin based combination therapy (ACT), is a serious global issue in malaria treatment and control. This warrants an urgent quest for novel compounds, particularly from natural sources such as medicinal plants. Alkaloids have over the years been recognized as important phytoconstituents with interesting biological properties. In fact, the first successful antimalarial drug was quinine, an alkaloid, which was extracted from Cinchona tree. In the present review work, the alkaloids isolated and reported recently (2013 till 2019) to possess antimalarial activity are presented. Several classes of alkaloids, including terpenoidal, indole, bisindole, quinolone, and isoquinoline alkaloids, were identified with a promising antimalarial activity. It is hoped that the reports of the review work will spur further research into the structural modification and/or development of the interesting compounds as novel antimalarial drugs.

Natural products have always played a significant role in the search for new drugs. One of the most relevant alkaloid-containing plant groups is the Amaryllidaceae family, a source of exclusive structures with a wide variety of pharmacological activities. The aim of this work was to determine the alkaloid composition and biological potential of an extract from the bulbs of an endemic Peruvian Amaryllidaceae species Ismene amancaes (Ker Gawl.) Herb. The alkaloid profiling was carried out by GC-MS, which revealed the presence of 13 compounds, 2 of them unidentified. The plant extract was found to contain high amounts of lycoramine, a galanthamine-type alkaloid. The extract also presented low inhibitory potential against the enzymes AChE and BuChE, with IC50 values of 14.6 ± 0.6 and 37.6 ± 1.4 μg·mL−1, respectively, and good to moderate inhibitory activity against the protozoan Plasmodium falciparum strain FCR-3 (chloroquine-resistant), with IC50 values of 3.78 ± 0.3 μg·mL−1. This is the first report of the alkaloid profile of a plant of the Ismene genus, which could be an interesting source of bioactive compounds.

There are no previous reports on the species under study, so this research constitutes the first report in this regard. In this way, the objective of the research ABSTRACT Introduction: The amaryllidaceae family is characterized by presenting alkaloids with powerful pharmacological activities, including antiprotozoal activity. The aim of the present work was to determine the chemical composition and evaluate the in vitro antiplasmodial activity of the total alkaloids of the bulbs of two amaryllidaceae species from northern Perú. Methods: The total alkaloids were extracted from the bulbs using an acid-base extraction. The chemical composition of the total alkaloids was determined by GC-MS, using galantamine as a reference standard. It was investigated the in vitro antiplasmodial activity against Plasmodium falciparum FCR-3 strain (chloroquine-resistant). Results: 8 alkaloids were identified in the bulbs of Clinanthus incarnatus: lycorine, galanthamine, galanthine, vittatine/crinine, hippamine, 3-O-acetylpowelline, 11,12-dehydroanhydrolycorine, 1-O-acetyllycorine with values of 19.73; 14.
