Effects of huperzine A on hippocampal inflammatory response and neurotrophic factors in aged rats after anesthesia

Abstract: Purpose: To investigate the effects of huperzine A (HupA) on hippocampal inflammatory response and neurotrophic factors in aged rats after anesthesia. Methods: Thirty-six Sprague Dawley rats (20-22 months old) were randomly divided into control, isofluran, and isoflurane+HupA groups; 12 rats in each group. The isoflurane+HupA group was intraperitoneally injected with 0.2 mg/kg of HupA. After 30 min, isoflurane inhalation anesthesia was performed in the isoflurane and isoflurane+HupA groups. After 24 h from anes… Show more

“…Antipsychotic drugs have been reported to attenuate POCD. Both galantamine and huperzine A are AChE inhibitors that reverse cognitive dysfunction by reducing the protein levels of IL-1β and normalizing excitatory synaptic transmission; huperzine A also increases the BDNF concentration ( Mao et al, 2016 ; Wang T. et al, 2019 ; Cai et al, 2020 ). Haloperidol is a dopamine receptor antagonist that reduces excessive dopaminergic activity during wakefulness, thereby reducing e.g., aggressive behavior ( Schrader et al, 2008 ; Nishigaki et al, 2019 ).…”

“…For instance, Lin and Zuo [3] have demonstrated increased expression of inflammatory cytokines in the hippocampus is probably involved in isoflurane-induced cell injury and cognitive impairment. Another study indicated that after the isoflurane anesthesia, the secretion of the hippocampal TNF-α, IL-6, and IL-1β levels was increased, while huperzine A rescued these expression levels [26]. These studies revealed that isoflurane could induce hippocampal inflammation and cognitive dysfunction.…”

“…Further study has shown that huperzine A could reduce neuronal degeneration in the CA1 region, and partially restore hippocampal choline acetyltransferase activity (Zhou et al 2001 ). In addition, huperzine A reduced the cognitive impairment of isoflurane anesthetized rats by inhibiting hippocampal inflammatory factors and increasing hippocampal neurotrophic factors (Cai et al 2020 ).…”
