Common bean (Phaseolus vulgaris L.) is an important legume with high nutritional value. In Nicaragua, certified healthy seeds of local bean varieties are not available, and seedborne fungi have gained little attention. Here, were surveyed seedborne pathogenic fungi in an important local bean cultivar, ‘INTA Rojo’. Beans grown in the four main production areas in Nicaragua (Boaco, Carazo, Estelí, Matagalpa) for future use as seed stock were sampled from four seed storehouses and six seed lots. A total of 133 fungal strains were isolated from surface-sterilized beans and inoculated to healthy lima beans (Phaseolus lunatus) under controlled conditions. Eighty-seven isolates caused symptoms of varying severity in the seedlings, including discoloration, necrotic lesions, cankers, rot, and lethal necrosis. Pathogenic isolates were divided into eight phenotypically distinguishable groups based on morphology and growth characteristics on artificial growth medium, and further identified by analysis of the internal transcribed spacer sequences (ITS1 and ITS2) of the ribosomal RNA genes. The pathogenic isolates belonged to eight genera. Fusarium spp. (F. chlamydosporum, F. equiseti, F. incarnatum), Lasiodiplodia theobromae, Macrophomina phaseolina, and Penicillium citrinum were the most damaging and common fungi found in the seed lots. Furthermore, Corynespora cassiicola, Colletotrichum capsisi, Colletotrichum gloeosporioides, Aspergillus flavus, and Diaporthe sp. (Phomopsis) were seedborne in cultivar ‘INTA Rojo’ and found to be pathogenic to bean seedlings. This study reveals, for the first time, many seedborne pathogenic fungi in beans in Nicaragua; furthermore, prior to this study, little information was available concerning F. equiseti, F. incarnatum, L. theobromae, C. cassiicola, and Diaporthe spp. as seedborne pathogens of common bean. Our results lay the basis for developing diagnostic tools for seed health inspection and for further study of the epidemiology, ecology, and control of the pathogenic fungi of common beans in the field.

This study investigated, at the microscopic level, whether the differential defence responses of soybean cultivars that are resistant (Fundacep 59) and susceptible (TMG 132) to target spot, caused by Corynespora cassiicola, could be associated with an increase in the production of phenolics, flavonoids and lignin at the infection sites. Many larger necrotic lesions with yellow halos were noticed on the leaves of plants from cultivar TMG 132, in contrast to the leaves of plants from cultivar Fundacep 59. Necrotic lesions also developed on the petioles of leaves of plants from cultivar TMG 132, while on the petioles and veins of leaves of plants from cultivar Fundacep 59, the lesions were of purple colour. The growth of fungal hyphae was reduced on the leaves of plants from cultivar Fundacep 59, and an apparently high density of trichomes was found in comparison with the leaves of plants from cultivar TMG 132. An appressorium‐like structure was produced at one or both extremities of the conidium of C. cassiicola, preferentially at the major and minor veins on the adaxial leaf surface of plants from both cultivars. Most cells on the leaves of plants from cultivar Fundacep 59 reacted against C. cassiicola infection by accumulating phenolic‐like compounds, which contributed to the death of many fungal hyphae and a greater maintenance of cell integrity. In contrast, fungal hyphae grew without any impedance in the leaf cells of plants from cultivar TMG 132, which was associated with signs of intense leaf tissue disorganization. Stronger autofluorescence and deposition of lignin and flavonoids were found in the cells of leaves of plants from cultivar Fundacep 59, in contrast to cultivar TMG 132. It can be concluded that soybean resistance to target spot is probably dependent on the activation of the phenylpropanoid pathway.

Considering the importance of target spot, caused by Corynespora cassiicola, to impact soya bean yield, this study investigated the C. cassiicola‐induced perturbations in the photosynthesis of soya bean plants from cultivars Fundacep 59 (moderately resistant) and TMG 132 (susceptible) by examining the chlorophyll (Chl) a fluorescence parameters and imaging and the photosynthetic pigment pools. The area under disease progress curve decreased by 38% for plants from cultivar Fundacep 59 in comparison with plants from cultivar TMG 132. The concentration of total Chla+b significantly increased by 16% and the Chla+b/carotenoids ratio significantly decreased by 11% for plants from cultivar Fundacep 59 in comparison with plants from cultivar TMG 132. Significant decreases of 31% and 8%, respectively, for the concentration of Chla+b and the Chla+b/carotenoids ratio occurred for inoculated plants in comparison with noninoculated ones regardless of the cultivar. As target spot developed, the values of maximal photosystem II quantum yield (Fv/Fm), effective photosystem II quantum yield [Y(II)], quantum yield of regulated energy dissipation [Y(NPQ)] and quantum yield of nonregulated energy dissipation [Y(NO)] increased, especially for plants from cultivar TMG 132. There were positive correlations of area under disease progress curve (AUDPC) with both Y(NPQ) and Y(NO) regardless of the cultivar. For cultivar TMG 132, the correlation between AUDPC and the electron transport rate was negative. Based on the analysis of the Chl a fluorescence, it was possible to elucidate the enormous potential of target spot to impair the functionality on the photosynthetic apparatus that may help the plants to invest in metabolic pathways involved in host defence responses.
