The influence of anaerobic protozoanBlastocystison human gut health is not well understood. WhileBlastocystisspecies frequently inhabit the gut, their clinical importance and ecological function remain ambiguous. A study onBlastocystiswas carried out enrolling a total of 203 participants including T2DM patients and non-diabetic individuals to evaluate the prevalence ofBlastocystisand its association in gut microbiota.Blastocystissubtypes were identified by PCR and faecal microbiome was accessed by targeting V4 region of the bacterial 16S ribosomal gene. The prevalence ofBlastocystisin T2DM was 25.49% and 17.82% in non-diabetic individuals with the most prevalent subtype on total population was ST3, followed by ST1 and ST2. The composition of gut microbiota was significantly different betweenBlastocystis-positive andBlastocystis-negative individuals.Blastocystiscarriage was positively associated with higher alpha diversity in T2DM patients and non-diabetic individuals. Interestingly, at the phylum level, the T2DM group had an obvious increase of Bacteroidetes and a marked increase of Actinobacteria with the present ofBlastocystis. The findings suggested that the presence ofBlastocystiswas linked to increased diversity and richness in the gut bacterial composition, signifying at a potentially beneficial association betweenBlastocystisand the gut microbiota.Author SummaryType 2 Diabetes Mellitus Patients (T2DM), a prevalent global disease, affects a significant portion of the population across the world. Thus, there is need to better understanding onBlastocystisinfection among T2DM that could lead to the alteration toward gut health. We evaluated the association betweenBlastocystisand gut microbiota, where involving two groups; T2DM patients and non-Diabetic individuals. The research revealed a higherBlastocystisin T2DM patients compared to non-diabetic individuals, emphasizing on assumption toward its pathogenicity. However, amplicon-based sequencing of 16S rRNA genes indicates thatBlastocystiscarriers exhibit increased gut microbiota diversity. Our result suggested that,Blastocystishighlighted its potential role as a component of a balanced microbiota. Notably, optimal alteration in Actinobacteria and Bacteroidetes may contribute to the several gut health. Hence, the study could prompt for further exploration regarding ofBlastocystissubtypes and gut microbiota specifically in T2DM to propose for more precise assessment ofBlastocystisand gut microbial diversity.

Prevalence and Molecular Characterisation of Blastocystis sp. Infecting Free-Ranging Primates in Colombia

Infection with Blastocystis sp. has been reported in free-living and captive non-human primates (NHPs); however, surveys on Blastocystis sp. from north-western South America are scarce. This study aimed to identify Blastocystis sp. in free-ranging NHPs living in Colombia. A total of 212 faecal samples were collected from Ateles hybridus, Cebus versicolor, Alouatta seniculus, Aotus griseimembra, Sapajus apella, and Saimiri cassiquiarensis. Smears and flotation were used for morphological identification. For samples microscopically classified as positive for Blastocystis sp., we used conventional PCR to amplify and sequence two regions of the SSU rRNA gene and used Maximum Likelihood methods and Median Joining Network analyses for phylogenetic analyses. Via microscopy, 64 samples were Blastocystis sp. positive. Through molecular analyses, 18 sequences of Blastocystis sp. subtype 8 (ST8) were obtained. Strain and allele assignment together with a comparative phylogenetic approach confirmed that the sequences were ST8. Alleles 21, 156, and 157 were detected. Median Joining network analyses showed one highly frequent haplotype shared by specimens from Colombia and Peru and close relationships between haplotypes circulating in NHPs from Colombia, Ecuador, Brazil, and Mexico. This survey could support the elaboration of a more accurate epidemiological picture of the Blastocystis sp. infecting NHPs.

Melo GB. Blastocystis sp.: molecular epidemiology and relationship with diseases in Brazilian scenarios. [thesis]. São Paulo: "Faculdade de Medicina, Universidade de São Paulo"; 2022. Blastocystis sp. is described as a protozoan prevalent in faecal samples from humans and animals, but still involves controversial aspects regarding its pathogenicity and epidemiology. In Brazil, there are few studies related to the understanding of relevant aspects of this enigmatic organism. Therefore, different approaches were proposed, such as the overview of research in Brazil in the last 20 years; the occurrence of Blastocystis sp. in the HC complex (Hospital das Clínicas) in the period of 2016 to 2021; the molecular characterization of isolates from patients with urticaria and diabetes, and perform in vitro cultivation of human isolates. The data of the literature review on the research of Blastocystis sp. in Brazil regarding the evaluation of its positivity in the HC-FMUSP complex reinforce the presence of this organism in our environment and raises the possibility of further studies to understand its real clinical significance. In relation to urticaria, it was not possible to establish a direct relationship with Blastocystis sp.Infection. Yet, it was possible to identify high intra and inter-subtypes diversity circulating in these patients. Regarding diabetics, although this protist was more prevalent in this group, it has not yet been possible to establish a relationship between diabetes and Blastocystis sp. Finally, our findings on the in vitro maintenance of human isolates allowed us to observed different cultivation times and forms present in different subtypes and alleles. Thus, further studies are needed focusing on the interaction between Blastocystis sp. and the host, to understand the impact of this organism on public health.
