Abstract:The basic knowledge on neoplasms is increasing quickly; however, few advances have been achieved in clinical therapy against tumors. For this reason, the development of alternative drugs is relevant in the attempt to improve prognosis and to increase patients' survival. Snake venoms are natural sources of bioactive substances with therapeutic potential. The objective of this work was to identify and characterize the antitumoral effect of Crotalus durissus terrificus venom (CV) and its polypeptide, crotoxin, on benign and malignant tumors, respectively, pituitary adenoma and glioblastoma. The results demonstrated that CV possess a powerful antitumoral effect on benign (pituitary adenoma) and malignant (glioblastoma multiforme) tumors with IC50 values of 0.96 ± 0.11 μg/mL and 2.15 ± 0.2 μg/mL, respectively. This antitumoral effect is cell-cycle-specific and dependent on extracellular calcium, an important factor for crotoxin phospholipase A2 activity. The CV antitumoral effect can be ascribed, at least partially, to the polypeptide crotoxin that also induced brain tumor cell death. In spite of the known CV nephrotoxicity and neurotoxicity, acute treatment with its antitumoral dose established in vitro was not found to be toxic to the analyzed animals. These results indicate the biotechnological potential of CV as a source of pharmaceutical templates for cancer therapy.

São Paulo: Instituto de Ciências Biomédicas da Universidade de São Paulo; 2011. A crotoxina é a toxina mais abundante e ativa do veneno da cascavel brasileira Crotalus durissus terrificus. Foi descrita como uma neurotoxina envolvida no bloqueio pré-sinaptico das junções neuromusculares. É uma proteína heterodimérica formada por uma unidade ácida (crotapotina) sem atividade enzimática, ligada à subunidade básica (PLA2) que possui de baixa toxicidade. No entanto, o heterodímero demonstra alta toxicidade, atribuída à interação entre a subunidade ácida que age como uma chaperonina e a básica que age como uma fosfolipase A2. Alguns autores atribuíram à esta toxina um efeito citotóxico e sugeriram seu uso como um agente terapêutico contra células tumorais. No presente trabalho, foi investigado o efeito citotóxico da crotoxina em células de melanoma murino (B16-F10) e fibroblastos (L929). As células foram tratadas com concentrações crescentes (partindo de 0,339 nM até 21,74 nM) de crotoxina por períodos de 6, 24 ou 48 horas. A viabilidade celular foi avaliada com ensaio de MTT. Estes ensaios foram repetidos com as subunidades da crotoxina em concentrações na proporção molar destas na molécula inteira. Os resultados dos ensaios de 48 horas mostraram que a crotoxina é altamente tóxica para as células de B16-F10 induzindo a morte celular até em concentrações mais baixas enquanto a linhagem L929 foi afetada somente em concentrações bem maiores. Os ensaios de 6 e 24 horas mostraram que o efeito citotóxico na linhagem B16-F10 foi verificado somente quando utilizaram-se as maiores concentrações da toxina, enquanto nenhuma morte celular foi observada na linhagem L929. Os ensaios com as subunidades demonstraram que a PLA2 é menos efetiva sem o carreador (crotapotina), e também que não houve efeito citotóxico nas células tratadas somente com a crotapotina. Estes resultados possibilitaram o cálculo da EC50% da crotoxina para ambas as linhagens sendo 0,08138 nM para a B16F10 e 6,91 nM para a L929. Palavras-chave: Crotoxina. Fosfolipase A2. Tumor. Neutralization of Bitis arietans venom-induced pathophysiological disorder, biological activities and genetic alterations by Moringa oleifera leaves

Bitis arietans venom (BAV) is known to cause various pathophysiological disorders by altering the cellular inclusions and enzymatic activities of different organs. Moringa oleifera leaf has been reportedly used for treatment of snake envenoming but there is no information on its neutralizing potentials against Bitis arietans venom. This study investigated the antivenin activity of ethanol crude extract of M. oleifera leaf on B. arietans envenomed rats and its inhibitory effects on some biological activities of the venom. The lethal dose (LD 50 ) of BAV was estimated at 1.5 mg/kg −1 . BAV induced various toxic effects in the in vivo study however, treatment with M. oleifera leaf extract (MOLE) ameliorated BAV-induced hypernatraemia and hypercalcemia. Acute anemia observed in untreated envenomed rats was reversed after treatment with various concentrations of MOLE with a significant (p < 0.05) dose dependent increase in hematological indices. Liver damage in untreated envenomed rats as indicated by higher concentration of serum liver enzymes and higher concentration of antioxidant enzymes were significantly (p < 0.05) decreased in MOLE treated rats. Also, BAV exhibited hemorrhagic, hemolytic and coagulating activities which were inhibited in a dose dependent manner by MOLE. A mild DNA fragmentation noticed in tissues of the heart of untreated envenomed rats was ameliorated in the MOLE treated rats. Results obtained in this study indicated that M. oleifera leaf have antivenin activity against B. arietans venom induced toxicities and underscores its use in folk medicine for the treatment of snake bites.
