Humoral immune response to antigens of Porphyromonas gingivalis ATCC 33277 in chronic periodontitis

Abstract: Introduction:Periodontitis is a chronic disease that results from an interaction of a mixed bacterial challenge and the host response. Objective:The purposes of this study were to evaluate the IgG serum levels to Porphyromonas gingivalis antigens by ELISA in individuals with different periodontal conditions correlated with clinical parameters, and to analyze the immunoreactivity profiles by Western blotting. Methods:Serum IgG levels against the cell sonicate antigen from P. gingivalis ATCC 33277 of 28 patients w… Show more

“…Another relevant result was similar production levels of IgG specific to P. gingivalis among the participants, regardless of whether they had severe asthma, periodontitis, or neither of these conditions. While the literature indicates that individuals with periodontitis are expected to harbor significantly higher levels of IgG specific to Porphyromonas gingivalis (17) , the present results were divergent. This would seem to suggest that the presence of severe asthma may influence the humoral response to Porphyromonas gingivalis, the relative quantities of Porphyromonas gingivalis were found to be significantly lower in the subgingival biofilm of individuals with chronicperiodontitis.…”

“…Confirming the role of Porphyromonas gingivalisin the etiology and pathogenesis of periodontitis, high levels of specific IgG against this bacteria were found in the sera of affected individuals (17,18) . Moreover, individuals with chronic periodontitis presented higher levels of IgG against Porphyromonas gingivalis antigens, such as HmuY and the gingipains proteinases, than healthy individuals, or those with gingivitis (17)(18)(19)(20)(21) .…”

“…27,28 In the present study, P. gingivalis infection showed significant increases in periodontal bone resorption in WT mice and TLR2 KO mice but not in TLR4 KO mice and TLR2&4 KO mice (Figure 2A-D), suggesting that TLR4 is essential for bone loss associated with P. gingivalis infection. However, RANKL protein levels were significantly higher in the P. gingivalis infection group in TLR4 KO mice (Figure 3G) and showed no significant change in TLR2&4 KO mice (Figure 3H), indicating that not only TLR4 signaling but also TLR2 signaling is involved in the RANKL-mediated bone loss.…”

“…33 The cells of P gingivalis, kindly provided by Dr P. Mastrantonio (Istituto Superiore di Sanità, Rome, Italy), were resuspended in PBS containing a protease inhibitor cocktail diluted 1:1000 (P-8465; Sigma-Aldrich) and subjected to ultrasonic disruption at output 3 in a sonifier (Bronson Sonic Power) for 20 minutes.…”
