Objetivo: Avaliar in vitro a ação antimicrobiana de materiais obturadores de canais radiculares de dentes decíduos por meio da difusão em ágar. Método: Os materiais testados foram: pasta Guedes-Pinto, pasta CTZ, OZE, Calen ® , L&C ® e MTA. Foi utilizada uma mistura microbiana composta por: Staphylococcus aureus-ATCC 6538, Enterococcus faecalis-ATCC 29212, Pseudomonas aeruginosa-ATCC 27853, Bacillus subtilis-ATCC 6633, Candida albicans-ATCC 10231. Foram utilizadas placas de Petri, contendo BHI ágar inoculadas com 0,1mL da mistura microbiana, nas quais foram confeccionadas cavidades no ágar, sendo estas preenchidas com as respectivas pastas. A leitura dos halos de inibição (mm) foi realizada após 48h/ 37ºC e os resultados submetidos aos testes Kruskal-Wallis e Mann-Whitney. Resultados: as pastas CTZ e Guedes-Pinto apresentaram halos de inibição estatisticamente maiores que os demais materiais (p<0,001). A L&C ® e o MTA não apresentaram halos de inibição. Conclusão: as pastas Guedes-Pinto, CTZ, Calen ® e o OZE apresentaram ação antimicrobiana por difusão. Dente decíduo; Tratamento do canal radicular; Microbiologia. Objective: To evaluate in vitro the antimicrobial action of root canal filling materials for primary teeth using the agar diffusion method. Method: The tested materials were Guedes-Pinto paste, CTZ paste, ZOE, Calen TM paste, L&C TM and MTA. A microbial mixture composed of the following microorganisms was used: Staphylococcus aureus-ATCC 6538, Enterococcus faecalis-ATCC 29212, Pseudomonas aeruginosa-ATCC 27853, Bacillus subtilis-ATCC 6633 and Candida albicans-ATCC 10231. Petri dishes containing BHI agar inoculated with 0.1 mL of the microbial mixture were used. After solidification of the culture medium, wells were prepared and immediately filled with the respective pastes. The diameter (mm) of the zones of microbial growth inhibition formed around the wells was measured after 48 h at 37ºC and the results were analyzed by the Kruskal-Wallis and Mann-Whitney tests. Results: The CTZ and Guedes-Pinto pastes presented significantly larger microbial growth inhibition zones than the other materials (p<0.001). L&C TM and MTA did not present inhibition zones. Conclusion: The CTZ, Guedes-Pinto and Calen TM pastes and ZOE cement presented antimicrobial action by diffusion. Deciduous tooth; Root canal therapy; Microbiology.

Antimicrobial analysis of different root canal filling pastes used in pediatric dentistry by two experimental methods

The objective of this study was to compare, by two experimental methods, the antimicrobial efficacy of different root canal filling pastes used in pediatric dentistry. The tested materials were: Guedes-Pinto paste (GPP), zinc oxide-eugenol paste (OZEP), calcium hydroxide paste (CHP), chloramphenicol + tetracycline + zinc oxide and eugenol paste (CTZP) and Vitapex. Fiven microbial strains (S. aureus, E. faecalis, P. aeruginosa, B. subtilis and C. albicans) obtained from the American Type Culture Collection were inoculated in Brain Heart Infusion (BHI) and incubated at 37 degrees C for 24 h. For the direct exposure test (DET), 72 paper points were contaminated with the standard microbial suspensions and exposed to the root canal filling pastes for 1, 24, 48 and 72 h. The points were immersed in Letheen Broth (LB), followed by incubation at 37 degrees C for 48 h. An inoculum of 0.1 mL obtained from LB was then transferred to 7 mL of BHI, under identical incubations conditions and the microbial growth was evaluated. The pastes showed activity between 1 and 24 h, depending on the material. For the agar diffusion test (ADT), 30 Petri plates with 20 mL of BHI agar were inoculated with 0.1 mL of the microbial suspension, using sterile swabs that were spread on the medium. Three cavities were made in each agar plate (total = 90) and completely filled with one of the filling root canal pastes. The plates were pre-incubated for 1 h at room temperature and then incubated at 37 degrees C for 24 to 48 h. The inhibition zone around each well was recorded in mm. The complete antimicrobial effect in the direct exposure test was observed after 24 h on all microbial indicators. All root canal filling materials induced the formation of inhibition zones, except for Vitapex (range, 6.0-39.0 mm).

BackgroundRecombinant amelogenin protein (RAP) is reported to induce complete root apex formation in dog model when used as apexification therapy. It also induces pulp regeneration in 85% of the treated group. Thus, the aim of this study was to investigate the nature of the remaining regenerated calcified tissues of the RAP group that showed no pulp regeneration compared to the calcium hydroxide treated group (CH).MethodsA total of 240 dogs’ open apex root canals were used, after establishment of canals contamination. Canals were cleaned, irrigated, and filled with RAP as an apexification material and compared with CH. Treated teeth were assessed by H&E, trichrome staining, and/or immunohistochemistry technique, at 1, 3, and 6 months.ResultsA time-dependent increase in the calcified tissue barrier was observed in the apex of the RAP-treated group compared to the CH-treated group. The newly formed dentin in this RAP group was mainly tubular dentin and was functionally attached to the bone by periodontal ligament, while the CH group showed dentin-associated mineralized tissue (DAMT) associated with the newly formed apical barrier.ConclusionsOut results suggest that RAP can be used as novel apexification material, resulting in a thickening and strengthening of the canal walls, and achieving apical closure.
