A genética em transformação: crise e revisão do conceito de gene

Abstract

O conceito de gene tem desempenhado um papel central na biologia desde sua introdução, no início do século xx. Contudo, ao longo do seu desenvolvimento histórico, o conceito tem sido objeto de controvérsia crescente, inicialmente na filosofia da biologia e, depois, na própria biologia. Desafios ao conceito de gene têm levado a uma dificuldade de preservar o chamado conceito molecular clássico, de acordo com o qual um gene é um segmento do DNA que codifica um produto funcional (polipeptídeo ou RNA). As últimas três décadas de estudos experimentais levaram a achados como genes interrompidos, emenda (splicing) alternativa, o chamado DNA-lixo, sequências TAR, pseudogenes, regulação pós-transcricional, rnai e rnasi, entre outros, os quais colocaram dificuldades inesperadas à compreensão usual do conceito de gene. Neste artigo, discutiremos os principais achados experimentais que desafiaram o conceito molecular clássico de gene. Daremos destaque, em particular, a avanços recentes, que tiveram lugar no Projeto Genoma Humano (PGH) e na Enciclopédia de Elementos de DNA (Encode). Atualmente, é clara a necessidade de uma análise e reformulação cuidadosa desse conceito central para o pensamento biológico. Muitos filósofos da biologia e biólogos, na tentativa de organizar a variedade de definições de gene, apresentaram visões interessantes a respeito desse conceito e de seu papel no conhecimento biológico, assim como propostas de revisão conceitual, que também abordaremos neste artigo. Concluímos que uma definição única de gene não é possível ou necessária. Ao contrário, o pluralismo de modelos e conceitos é provavelmente mais poderoso, desde que os domínios de cada conceito ou modelo sejam claramente definidos.<br>The gene concept has played a central role in Biology since its introduction, in the beginnings of the twentieth century. However, throughout its historical development, this concept has been a matter of increasing controversy, initially in the philosophy of biology and, later, in biology itself. Challenges to the gene concept have resulted in the difficulty of preserving the so called classical molecular concept, according to which a gene is a stretch of DNA encoding a functional product (polypeptide or RNA). The last three decades of experimental studies led to findings such as interrupted genes, alternative splicing, so called junk DNA, TAR sequences, pseudogenes, postranscriptional regulation, RNAi and RNAsi, among others, that posed unexpected difficulties to the usual understanding of the gene concept. In this paper, we address the main experimental findings that challenge the classical molecular gene concept. We focus, in particular, on recent advances that took place in the Human Genome Project (hgp) and the Encyclopedia of DNA elements (Encode).
