Expression pattern of glycoconjugates in the Bidderian and ovarian follicles of the Brazilian toad Bufo ictericus analyzed by lectin histochemistry

Abstract: The Bidder's organ and ovary of the Brazilian toad Bufo ictericus were studied by light microscopy, using hematoxylin-eosin (HE) and periodic acid Schiff (PAS) staining. The expression and distribution of carbohydrate moieties was analyzed by lectin histochemistry, using 8 lectins with different carbohydrate specificities: Ulex europaeus (UEA I), Lens culinaris (LCA), Erythrina cristagalli (ECA), Arachis hypogaea (PNA), Ricinus communis (RCA I), Aleuria aurantia (AAA), Triticum vulgaris (WGA), and Glycine maxi… Show more

“…When compared to Bombina pachypus , which we studied previously (Scillitani et al, ), B. bufo and B. balearicus have a lower number of jelly layers, but a comparable number of glycan types as evidenced by lectin histochemistry. Similarly, a survey of the available literature about ECM composition in bufonid toads (Arranz et al, ; Coppin et al, ; Farias et al, ; Kawai and Anno, ; Morelle et al, ; Omata, ; Reddy and Seshadri, ) indicates that in the two species we studied there are the main residuals found in other species, such as galactose, N ‐acetylgalactosamine, N ‐acetylglucosamine, sialic acid, fucose, and mannose. Besides, patterns of lectin binding allow to discriminate the two species between them and could be even used as a tool for egg identification.…”

“…), including the eight globo series glycans, blood A antigen and α -Gal. For cross comparison studies with VK9, we select two additional binding proteins. The lectin Helix pomatia agglutinin (HPA) has specificity towards the terminal α GalNAc of A glycan[40][41][42], while Erythrina Cristagalli Lectin (ECA) has specificity towards the terminal lactose of Gb2 [43]. Three multivalent glycan-DNA conjugates and their specific GBPs (GbH vs VK9, blood A antigen vs HPA, and Gb2 vs ECA) were subjected to singleplex selection with different quantity (10, 5, 1, 0.1 pmol of conjugates), followed by qPCR assay.…”
