Staphylococcus aureus is a pathogen that frequently causes mastitis in bovine herds worldwide. This pathogen produces several virulence factors, including cell-associated adhesins, toxic and cytolytic exoproteins, and capsular polysaccharides. The aim of the present study was to test for the presence of genes involved in capsular polysaccharide production and biofilm formation in S. aureus isolated from bovine mastitis samples collected from 119 dairy herds located in three different Brazilian regions, as well as to assay the production of capsular polysaccharides and biofilm, in vitro. The detection of the cap, icaAD, and bap genes was performed using PCR. The detection and quantification of capsular polysaccharide production was performed using ELISA assays. The ability of the isolates to form a biofilm was examined using the polystyrene surface of microtiter plates. All 159 S. aureus isolates investigated harboured the cap gene: 80 % carried the cap5 gene and 20 % carried the cap8 gene. Sixty-nine percent of the isolates expressed capsular polysaccharide (CP) in vitro, 58 % expressed CP5 and 11 % expressed CP8. All of the isolates harboured the icaA and icaD genes, and 95.6 % of the isolates carried the bap gene. Of the 159 isolates analysed, 97.5 % were biofilm producers. A significant association between the capsular genotype and phenotype and the amount of biofilm formation was detected: cap5/CP5 isolates tended to form more biofilm and to produce a thinner CP layer than cap8/CP8 isolates. The results indicate a high potential for pathogenicity among S. aureus isolated from bovine milk collected from three different regions in Brazil.

Staphylococcus spp. can survive in biofilms for long periods of time, and they can be transferred from one point to another and cause environmental contamination in food processing. The aim of this study was to detect Staphylococcus strains isolated from a poultry processing plant by the presence of adhesion genes and the phenotypic production of exopolysaccharide. In the present study, the production of exopolysaccharide and the presence of adhesion genes in 65 strains of Staphylococcus spp. were evaluated. All strains of Staphylococcus spp. produced exopolysaccharide, as confirmed by formation of black and opaque colonies in Congo Red Agar. The variation of sucrose content was critical for the production of exopolysaccharide in Congo Red Agar since at low sucrose concentrations all strains presented a characteristic result, i.e., there was no exopolysaccharide production. The atl gene was found in all strains, and the icaA and icaD genes were found in 97% of them. The data obtained suggest that Staphylococcus spp. isolated from the poultry processing plant evaluated has a potential for biofilm formation. An efficient control of this microorganism in food processing environment is necessary as they may represent a potential risk to consumers.

The Candida parapsilosis complex comprises three genetically related species: Candida parapsilosis sensu stricto, Candida orthopsilosis and Candida metapsilosis. During the last decade, Candida parapsilosis sensu stricto has become the second most commonly isolated yeast from patients with blood stream invasive fungal infections, following Candida albicans. Despite its clinical significance, little is known about its distribution and behavior in oral cavity niches. Given this background, we intended to perform a molecular characterization of 38 isolates of the Candida parapsilosis complex collected from oral cavities of immunocompetent patients with and without gingivalperiodontal disease, with the aim of determining its distribution and establishing a correlation with clinical data. Besides, we assessed the virulence of the strains by examining the in vitro biofilm-forming capacity in two different culture media: YPD and RPMI 1640. Being more frequently isolated from oral cavities with pathological conditions, 89.5% of the strains were reconfirmed to be Candida parapsilosis sensu stricto. More than 70% of the Candida parapsilosis sensu stricto strains showed biofilm-forming capacity in both culture media, reflecting a statistically significant association between biofilm-forming capacity and the clinical conditions of the subjects from whom the strains were isolated. Among the three complex species, Candida parapsilosis sensu stricto is a normal commensal in the oral cavity, being more frequently recovered in inflammatory conditions. Gingival-periodontal disease may be a factor triggering overgrowth and virulence of Candida parapsilosis sensu stricto commensal strains.
