Background: MicroRNAs (miRNAs) play an essential role in gene regulators in many biological and molecular phenomena. Unraveling the involvement of miRNA as a key cellular factor during in vitro canine parvovirus (CPV) infection may facilitate the discovery of potential intervention candidates. However, the examination of miRNA expression profiles in CPV in tissue culture systems has not been fully elucidated. Method: In the present study, we utilized high-throughput small RNA-seq (sRNAseq) technology to investigate the altered miRNA profiling in miRNA libraries from uninfected (Control) and CPV-2c infected Crandell Reese Feline Kidney cells. Results: We identified five of known miRNAs (miR-222-5p, miR-365-2-5p, miR-1247-3p, miR-322-5p and miR-361-3p) and three novel miRNAs (Novel 137, Novel 141 and Novel 102) by sRNA-seq with differentially expressed genes in the miRNA repertoire of CPV-infected cells over control. We further predicted the potential target genes of the aforementioned miRNAs using sequence homology algorithms. Notably, the targets of miR-1247-3p exhibited a potential function associated with cellular defense and humoral response to CPV. To extend the probing scheme for gene targets of miR-1247-3p, we explored and performed Gene Ontology (GO) enrichment analysis of its target genes. We discovered 229 putative targets from a total of 38 enriched GO terms. The top over-represented GO enrichment in biological process were lymphocyte activation and differentiation, marginal zone B cell differentiation, negative regulation of cytokine production, negative regulation of programed cell death, and negative regulation of signaling. El objetivo del estudio fue la detección de parvovirus canino tipo 2 (CPV-2) en perros jóvenes con/sin sintomatología clínica compatible con parvovirosis mediante la técnica de PCR, usando cebadores que pueden permitir la amplificación de un fragmento del gen codificante de la proteína VP2. Se colectaron hisopados rectales de 78 perros menores a un año y sin historia de vacunaciones previas, de los cuales 39 individuos tuvieron un diagnóstico clínico de parvovirosis canina y los otros 39 fueron animales clínicamente sanos. Para la extracción de ADN viral se usó el método fast boiling, donde las muestras fueron sometidas a un hervido a 100 °C por 10 minutos con posterior centrifugación para extraer el sobrenadante, el cual fue usado como molde para la reacción de PCR. Se usaron cebadores específicos que amplifican un fragmento de 1316 pares de bases del gen VP2 del virus CPV-2, utilizando como control positivo una vacuna comercial. El virus fue detectado en el 62% de animales con diagnóstico clínico de la enfermedad con PCR convencional, no siendo detectado en perros clínicamente sanos. La no detección de CPV-2 en animales con diagnóstico clínico compatibles a parvovirosis en el 38% de los casos indicaría la presencia de otro agente etiológico como causante del cuadro sintomatológico, recomendándose el uso de técnicas complementarias para el correcto diagnóstico de la enfermedad.

The aim of the present study was to detect canine parvovirus (CPV) from faecal samples of clinically ill domestic dogs by polymerase chain reaction (PCR) followed by VP2 gene partial sequencing and molecular characterization of circulating strains in Lithuania.Eleven clinically and antigen-tested positive dog faecal samples, collected during the period of 2014-2015, were investigated by using PCR. The phylogenetic investigations indicated that the Lithuanian CPV VP2 partial sequences (3025-3706 cds) were closely related and showed 99.0-99.9% identity. All Lithuanian sequences were associated with one phylogroup, but grouped in different clusters. Ten of investigated Lithuanian CPV VP2 sequences were closely associated with CPV 2a antigenic variant (99.4% nt identity). Five CPV VP2 sequences from Lithuania were related to CPV-2a, but were rather divergent (6.8 nt differences). Only one CPV VP2 sequence from Lithuania was associated (99.3% nt identity) with CPV-2b VP2 sequences from France, Italy, USA and Korea. The four of eleven investigated Lithuanian dogs with CPV infection symptoms were vaccinated with CPV-2 vaccine, but their VP2 sequences were phylogenetically distantly associated with CPV vaccine strains VP2 sequences (11.5-15.8 nt differences).Ten Lithuanian CPV VP2 sequences had monophyletic relations among the close geographically associated samples, but five of them were rather divergent (1.0% less sequence similarity). The one Lithuanian CPV VP2 sequence was closely related with CPV-2b antigenic variant. All the Lithuanian CPV VP2 partial sequences were conservative and phylogenetically low associated with most commonly used CPV vaccine strains.
