Isolation of the first IMP-4 metallo-β-lactamase producing Klebsiella pneumoniae in Tianjin, China

Abstract: This study shows for the first time the mechanism of carbapenem resistance of a Klebsiella pneumoniae clinical isolate TJ8 recovered from Tianjin Medical University General Hospital ,China. The modified Hodge test and EDTA synergy test were performed for the screening of carbapenemases and metallo-β-lactamases (MBLs), respectively. “…PCR was carried out in 25 μL reactions using 12.5 μL of PCR master mix (Nippon genetics, Europe), 1 μL (12.5 pmol) of each primer, 2μL (20ng) of template DNA and 8.5 μL of sterile water. The amplification reactions were carried out by conventional PCR using Biometra, UNO II thermal cycler under the following conditions: for blaIMP-1, blaIMP-2 and blaVIM-1; initial denaturation at 94°C for 5 minutes, followed by 35 cycles each consists of denaturation at 94°C for 30 seconds, annealing at 55°C for 30 seconds, extension at 72 °C for 90 seconds with a final extension step at 72°C for 10 minutes; for blaVIM-2 gene the same conditions were used except that the initial denaturation was for 3 minutes [12,13]. The resulting products were loaded in 1.5% agarose gel and were visualized under UV illumination system.…”

“…MBL-producing K. pneumoniae isolates have usually been reported sporadically in China (Mendes et al 2008;Liu et al 2009;Li et al 2012;Wang et al 2014), and some researches have demonstrated, at most, a low-level endemicity of MBL producers in hospitals where similar strains have been detected (Chen et al 2011;Liu et al 2014).
