Intensive fish farming systems in Brazil have increased the disease incidence, mainly of bacterial origin, due to higher stocking density, high organic matter levels and poor quality of the aquatic environment that causes high mortality rates during outbreaks. The identification of pathogenic species using a fast and reliable method of diagnosis is essential for successful epidemiological studies and disease control. The present study evaluated the use of direct colony PCR in combination with 16S rRNA gene sequencing to diagnose fish bacterial diseases, with the goal of reducing the costs and time necessary for bacterial identification. The method was successful for all 178 isolates tested and produced bands with the same intensity as the standard PCR performed using pure DNA. In conclusion, the genetics methods allowed detecting the most common and important pathogens in Aquaculture, including 12 species of occurrence in Brazilian fish farms. The results of the present study constitute an advance in the available diagnostic methods for bacterial pathogens in fish farms.

Isolation and molecular identification of the etiological agents of streptococcosis in Nile tilapia (Oreochromis niloticus) cultured in net cages in Lake Sentani, Papua, Indonesia

Infections with Streptococcus spp. were observed in Nile tilapia cultured in net cages in Lake Sentani, Papua, Indonesia. Clinical signs included exophthalmia, erratic swimming, ascites in abdominal cavity, and external hemorrhages. Four types of bacterial colonies (SK, K10, P20, and M12) were isolated from the brain, kidney, and eyes. Based on phenotypic and genetic (16S rDNA sequencing) characteristics, the isolates were identified as Streptococcus iniae (SK), Streptococcus agalactiae (K10 and P20) and Lactococcus garvieae (M12). The latter species has not been previously isolated or reported from fish streptococcosis in Indonesia. Intraperitoneal injection of healthy tilapia with the bacterial species caused significant morbidity (70%) within 3 days and 100% mortality at 6 days post injection. Experimental infections and reisolation of the bacteria from morbid and dead fish suggest they are the causative agents of streptococcosis, which rendered high mortality among cage cultured Nile tilapia in Lake Sentani. Our results suggest the need for developing diagnostic tools for accurate identification of the agents of streptococcosis. As tilapia aquaculture continues to expand as a means of food production and livelihood in Indonesia, it becomes crucial to ensure that fish resources are monitored and protected from the adverse effects of infectious diseases.

Objective. This investigation aimed to study the presence of Streptococcus spp. in tilapia (Oreochromis niloticus) from fish farm located in Sullana-Piura, Peru. Materials and methods. 150 fish with clinical signs of streptococcal disease were sampled, and the bacterium isolation was performed on blood agar, correlated to histopathological lesions description and molecular confirmation by real-time PCR. Results. The necropsy revealed exophthalmia, hyphema, congestion and/or haemorrhagic meninges, ascites, splenomegaly, hepatomegaly and diffuse haemorrhagic zones throughout the body. 102 isolated positives (54 tilapias) to Streptococcus spp. were identified in the microbiological analysis (prevalence of 26%), the brain was the organ with the highest percentage of this bacteria (34.31%), and 19 isolates were beta-haemolytic (18.63%) with prevalence of 10.12%. Fish beta-haemolytic streptococci presented epicarditis, perisplenitis and chronic meningitis, panophthalmitis, coagulative necrosis of skeletal muscle and granulomas formation. In the confirmatory test by real-time PCR, any positive tilapia to S. iniae was obtained. The results were analysed using a stochastic simulation of beta distribution using @Risk program uncertainty, reporting an average prevalence of 0.66% in sick tilapias. Conclusions. The analysed fishes were positive to bacteria of the genus Streptococcus, which confirms its presence in the fish farm. However, 19 isolates were beta-haemolytic, and the presence of S. iniae was not positive to the limit prevalence of 2.7% in real-time PCR.Keywords: Cichlids, histopathology, microbiology, PCR, teleost fish (Source: DeCS, CAB, MeSH).
