Evaluating most probable number method to count and isolate viable methylotrophs

Abstract: Nine different receptacles were tested with the MPN method to determine which receptacle was most reliable and economical for MPN counts. Results showed that 96 well PCR plate were the best vessels for this type of analysis and facilitated the isolation of viable Methylotrophs.

“…In this way, the modified MPN drop agar method provides significant benefit compared to current state of art [11,[22][23][24][25], compared especially with drop agar methods cited in the literature. There, 96-well tissue plate was obviously used for dilution preparation and drops with dilutions were inoculated to one plate [26][27][28].…”

“…The MPN method is one of the primary methods for the enumeration of L. monocytogenes and other foodborne pathogens, and is supported by enrichment schemes that can potentially suppress the level of background flora (Capita and Alonso-Calleja, 2003;Chen et al, 2017). However, the MPN method is laborious and requires the use of a large amount of media (Sutton, 2010;Kashyap, 2011;Erkmen and Bozoglu, 2016). Furthermore, the MPN method cannot be applied for a large number of samples simultaneously, making it difficult to utilize the data for constructing a growth curve under various conditions (Kawasaki et al, 2014).…”
