Principles and applications of polymerase chain reaction in medical diagnostic fields: a review

Abstract: Recent developments in molecular methods have revolutionized the detection and characterization of microorganisms in a broad range of medical diagnostic fields, including virology, mycology, parasitology, microbiology and dentistry. Among these methods, Polymerase Chain Reaction (PCR) has generated great benefits and allowed scientific advancements. PCR is an excellent technique for the rapid detection of pathogens, including those difficult to culture. Along with conventional PCR techniques, Real-Time PCR has… Show more

“…1 This standard amplification technique can be applied for various biological studies such as genome sequencing, molecular genetic analysis, diagnosis of infectious disease, and genetic disorder. 2,3 Even though PCR is an efficient gene-amplification system, the expensive device, requirement of a trained person, long turnaround times, non portability, and difficulties in the quantification of end products are considered to be limitations of the real-time diagnosis field. 4 As a result, real-time PCR and quantitative PCR systems have been developed to both amplify and detect a DNA simultaneously.…”

“…Polymerase chain reaction (PCR) offers a fast, reliable, and economical alternative to histology and culture, and is quickly becoming the most widely used method for detecting human, animal, and plant pathogens. 11,15,19,21,23 The current study describes a newly developed PCR-based test to detect the WNS-associated fungus G. destructans directly from bat wing tissue within hours. Reliability of the PCR method was assessed by comparisons with culture and histology techniques.…”
