tfraso@usp.br Birds of the Cracidae family (curassows, guans, and chachalacas) are endemic of the Neotropics and 50 species are currently classified. Brazil has 22 species, seven of which are considered threatened. The Alagoas Curassow (Pauxi mitu) species is considered extinct in the wild; but about 120 birds are alive in captivity. Conservation of this species depends entirely on correct management. Health reports of both wildlife and captive curassows are rare. In this study the presence of Escherichia coli was evaluated in 23 healthy Alagoas Curassows from two private breeding centres. E. coli was isolated from cloacal swabs, and the presence of genes encoding cytotoxic necrotising factor 1 (cnf1), alpha-haemolysin (hly), aerobactin (iuc), serum resistance (iss) and the following adhesions: S fimbriae (sfa), pili associated with pyelonephritis (pap) and temperature-sensitive haemagglutinin (tsh) were investigated. E. coli was isolated from 78.3% (18/23) of the birds, and the percentage of curassows colonized by E. coli was similar between the two facilities. From the 22 E. coli isolates, 15 (68.2%) were positive for at least one virulence factor by PCR, and the most frequently found gene was iss (50%). No curassows had clinical signs of disease. Nevertheless, the presence of some E. coli strains may be a concern to the wildlife in captivity. Problem statement: Omphalitis is one of the most common causes of mortality in chicks during the first week after hatching. Escherichia coli strains are the most common isolated bacteria from omphalitis cases of chickens. Bacterial colonization in the host cells surfaces is a critical first step in the pathogenesis of avian pathogenic Escherichia coli isolates. Thus the current study was undertaken to determine the presence and prevalence of several adhesin-encoding genes in E. coli isolates from omphalitis of chicks. Approach: One hundred four E. coli isolates were recovered from omphalitis cases and were identified by standard biochemical tests. The omphalitis-derived isolates were examined for the presence of fimbrial and non-fimbrial adhesin-encoding genes by PCR technique. Results: Most (93.26%) of the E. coli isolates exhibited at least one of the examined adhesin-encoding genes. None of the isolates contained the afaI B-C, afa E-VIII and f17A genes. The two most prevalent genes were crl (87.50%) andfimH (77.88%). P (papC) and S (sfa) fimbriae encoding genes were detected in 8 (7.69%) and 5 (4.80%) isolates respectively. Seven combination patterns of the adhesin-encoding genes were detected. In 83 (79.80%) isolates combinations of 2-4 genes were detected. The gene combinations of crl-fimHand fimH-papC were the two most prevalent patterns respectively. Fourteen (13.46%) isolates showed crl gene alone and 7 (6.73%) isolates were negative for examined genes. Conclusion: The current study showed that some of the adhesin-encoding genes are more prevalent in E. coli isolates from omphalitis of chicks but, E. coli isolates may be expressing still unknown adhesins that could have a role in the pathogenicity of omphalitis-derived isolates

The objective of this study was to determine the inhibition ability of the earthworm (L. rubellus) extract (ECT), dried earthworm extract (ECT-k), and granule earthworm extract (ECT-g) as poultry feed additive against some pathogenic bacteria. Antibacterial activity was performed using diffusion method against Escherichia coli, Salmonella pullorum, Pseudomonas aeruginosa, and Staphylococcus aureus in vitro. In the present study, the concentrations of ECT, ECT-k, and ECT-g in nutrient broth (NB) media tested were consisted of treatments A: 0%, B: 0.26%, C: 0.52%, D: 0.78% and E: 1.04% (g/vol) respectively. The results of the in vitro study showed that started from ECT level 0.26% inhibited (P <0.05) growth of P. aeruginosa and S. aureus, while ECT level 0.52% inhibited (P <0.05) E. coli and S. pullorum which proportional to the increased in concentration. ECT-k level 0.26% inhibited (P <0.05) growth of E. coli and S. aureus, while ECT-k level 0.52% inhibited (P <0.05) P. aeruginosa, and ECT-k level 1.04% inhibited (P<0.05) growth of S. pullorum. ECT-g level 0.26% inhibited (P <0.05) growth of S. pulorum, while ECT-g level 0.52% inhibited (P <0.05) S. aureus and ECT-g level 1.04% inhibited (P<0.05) growth of P. aeruginosa. There were no antibacterial action (P>0.05) of ECT and ECT-t against S. pullorum. Diameter of inhibition zone for 24 hours showed that S. aureus was the most sensitive bacterium to ECT and ECT-k, and S. pullorum was the most sensitive bacterium to ECT-g.
