In the past two decades the members of the genus Enterococcus have emerged as important nosocomial pathogens worldwide. In the present study, we evaluated the antimicrobial resistance and genotypic characteristics of 203 Enterococcus spp. recovered from different clinical sources from two hospitals in Porto Alegre, Rio Grande do Sul, Brazil. The species were identified by conventional biochemical tests and by an automated system. The genetic diversity of E. faecalis presenting high-level aminoglycoside resistance (HLAR) was assessed by pulsed-field gel electrophoresis of chromosomal DNA after SmaI digestion. The E. faecalis was the most frequent specie (93.6%), followed by E. faecium (4.4%). The antimicrobial resistance profile was: 2.5% to ampicillin, 0.5% to vancomycin, 0.5% teicoplanin, 33% to chloramphenicol, 2% to nitrofurantoin, 66.1% to erythromycin, 66.5% to tetracycline, 24.6% to rifampicin, 30% to ciprofloxacin and 87.2% to quinupristin-dalfopristin. A total of 10.3% of the isolates proved to be HLAR to both gentamicin and streptomycin (HLR-ST/GE), with 23.6% resistant only to gentamicin (HLR-GE) and 37.4% only to streptomycin (HLR-ST). One predominant clonal group was found among E. faecalis HLR-GE/ST. The prevalence of resistance among beta-lactam antibiotics and glycopeptides was very low. However, in this study there was an increased number of HLR Enterococcus which may be spreading intra and inter-hospital.

Three Enterococcus faecalis and one Enterococcus faecium strains were characterized by plasmid profile, pulsed-field gel electrophoresis (PFGE) and determination of antimicrobial minimal inhibitory concentrations. VanA elements were characterized by Long PCR, overlapping PCR and DNA sequencing. Enterococcal strains showed resistance to vancomycin and harbored the vanA gene, and three these were teicoplanin susceptible while one showed intermediate resistance to teicoplanin. Two E. faecalis strains showed indistinguishable PFGE profile while the third was unrelated. E. faecalis strains showed a deletion in the right terminal region of the Tn1546-like element. The E. faecium strain showed an insertion element in the vanXY intergenic region. Mutations in VanA elements were not found. Rearrangements in the VanA element could be responsible for incongruities in genotype and phenotype in these strains.

Effect of Spirulina platensis Powder on Whey Fermented with Streptococci

The main purpose of this research was to monitor the effect of dry Spirulina platensis powder in whey inoculated with pure culture of Streptococcus lactis NCIM 2080 and Streptococcus faecalis NCIM 2093, respectively during their 21 days storage at 4°C. The whey samples were supplemented with 0.3, 0.5 and 0.8% w/v S. platensis powder and without S. platensis powder (control). All samples were inoculated with 1% v/v single strain of streptococci. All samples were inoculated with same viable counts. Whey samples were filled in 50 mL tightly capped sterilized falcon tubes. Five hours of incubation were given to all samples each in three replicates at 40°C. The effect was studied on following parameters: pH, titratable acidity and redox potential of whey and viability of streptococci in current sample. These attributes were checked on 0th day (before incubation), 1st, 7th, 14th and 21st day of storage. The most significant results were obtained at 0.8% S. platensis supplemented samples. At the day 7 both streptococci gave the highest viable counts throughout the 21 days of storage. The lowest pH and highest titratable acidity was observed in S. lactis at 21st day of storage.
