Production of glucosyltransferase by Erwinia sp. using experimental design and response surface methodology

Abstract: Glucosyltransferase produced by strain Erwinia sp. is an intracellular enzyme that catalyzes the formation of isomaltulose from sucrose. Isomaltulose is a non-cariogenic reducing dissacharide commercially used in foods. Response surface methodology and 2 3 -factorial central composite design were employed to optimize a fermentation medium for the production of glucosyltransferase by Erwinia sp. in shaken flasks at 200 rpm and 30ºC. The three variables involved in this study were sugar cane molasses (SCM), corn… Show more

“…Central composite design (CCD) is a well established, widely used statistical technique for determining the key factors from a large number of medium components by a small number of experiments. Experimental designs have been commonly used for the optimization of multiple variables with minimum number of experiments (10,13,15,19,26). In this investigation, we tried to improve cell growth as well as nitrilase production of recombinant E. coli by studying various medium components using central composite design.…”

“…Response surface method (RSM) is a widely used technique for optimization of fermentation process parameters. RSM, which is based on Box-Behnken's design, has advantages that include fewer experiments, suitability for multiple factor experiments, search for relativity between factors, and finding of the most suitable condition and prediction of response [15,16]. In this method, linear or quadratic modelings of experimental variables generate contour plots and a model equation fitting the experimental data.…”
