Optimization of Fermentation Medium for Enhanced Glucansucrase and Glucan Production from Weissella confusa

Abstract: 18.2, 18.0, 7.0, 6.4, 6.2 and 6.4 U/ml enzyme activity and 103, 100, 46, 41, 39 and 37 mg/ml glucan concentration, respectively. Sodium acetate and beef extract were not effective for enzyme and glucan production. The new strain Weissella confusa can be used for commercial production of glucansucrase and glucan using optimized medium.

“…The similar results can also be found for other dextranproducing strains such as NRRL B-640 in research of Purama and Goyal [29]. The lower concentration of sucrose (5%) in the growth medium was the optimal for DS production by Weissella confusa Cab3 [30] and Lactobacillus plantarum DM5 [31].…”

“…The optimum pH for DS production from our strain is in correlation with other reports for glucansucrases from various lactic acid bacteria viz. L. mesenteroides NRRL B-640 [29], W. confusa Cab3 [30] and Lb. plantarum DM5 [31].…”

“…Weissella confusa Cab3 grew well within the range of pH 5.0 to 7.0, and temperature 20˚C to 45˚C however Weissella confusa Cab3 produced maximum dextransucrase (6.0 U/ml) at pH 7.0 and 25˚C in the enzyme production medium [19]. As reported earlier, the effects of several medium components were investigated and sucrose, Tween80, yeast extract and K 2 HPO 4 were effective nutrients which displayed higher dextransucrase production [20]. Based on the earlier results, L-16 Taguchi's orthogonal array method was designed, where the factors were varied in four levels for determining optimal medium components for dextransucrase production from Weissella confusa Cab3.…”

“…To achieve the maximum dextransucrase production by Weissella confusa Cab3, sucrose (%, w/v), Yeast extract (%, w/v), dipotassium hydrogen orthophosphate (K 2 HPO 4 ; %, w/v) and Tween80 (%, v/v) were selected for medium optimization for enhanced dextransucrase production because they had significant impact on dextransucrase production as screened in our earlier findings [20]. Sucrose was the most effective medium component for dextransucrase production.…”
