Cryopreservation of semen from functional sex-reversed genotypic females of the rainbow trout, Oncorhynchus mykiss

Abstract: Cryopreservation of semen from sex-reversed females of rainbow trout aims at rationalizing the production of stocks composed by 100% females. Semen from normal males (M) and two types of genotypic females (R and G), sex-reversed by the oral administration of 17α-methyltestosterone, were used. R was obtained by the fertilization of normal eggs with semen of sex-reversed females while G via gynogenetic reproduction. Semen was diluted in anextender solution (glucose 5,4 g, egg yolk 10 ml, dimetil sulfoxide 10 ml,… Show more

“…For this reason, a high rate of thawing is usually used in order to diminish the level of recrystallization. When the thawing is rapid, there is not enough time for the dehydrated cells to absorb the quantity of water the lost during freezing (Ninhaus-Silveira et al, 2006a;NinhausSilveira et al, 2006b). In March / 2004, it was reported that a baby was born from semen frozen for 21 years and sent to the in vitro fertilization technique (Horne et al, 2004).…”

“…It also needs to be considered that steroids concentrations may decrease with storage time, especially at room temperature [ 21 , 40 ]. Optimising the dose and duration of treatment with masculinising agents is of the utmost importance; too-low concentrations may be ineffective (however, it may be useful for the production of functional males [ 38 ]), whereas too high doses often have a so called “paradoxical feminisation” or a sterilising effect [ 1 , 41 , 42 ].…”

“…The first attempt was conducted by [ 12 ] who cryopreserved the semen of SRF Chinook salmon in pellets using 0.3 M glucose and 10% DMSO and obtained fertilisation rates of about 80%. The use of an extender consisting of glucose, DMSO, and egg yolk led to the fertilising ability of the cryopreserved sperm of SRF rainbow trout being about 60% [ 24 , 38 ]. The cryopreservation of SRF rainbow trout semen can be also performed using multicomponent mineral solution of [ 123 ] supplemented with only DMSO [ 106 ] or DMSO, egg yolk, and Dan Pro S760 [ 102 ].…”
