Effects of plant lectins on in vitro fibroblast proliferation

Abstract: Lectins are carbohydrate-binding proteins that have been isolated from various sources and presented a wide spectrum of biological activities. The effects of four lectins, namely, Phaseolus vulgaris phytohemagglutinin, PHA, wheat germ agglutinin, WGA, Artocarpus integrifolia seed lectins, jacalin and artocarpin, on in vitro fibroblasts proliferation were investigated. The lectins did not influence the initial cell adhesion to the plate. PHA and WGA at 10-20 µg/mL concentrations significantly decreased fibrobla… Show more

“…Several studies have confirmed the use of lectins in the immune system activation, enlisting neutrophils through indirect mechanisms [ 41 ], promoting proinflammatory effects in polymorphonuclear cells and inducing the cytokines release [ 42 ], as well as triggering fibroblasts proliferation [ 43 ]. Previous assays accomplished by our group have shown a potential proinflammatory and immunomodulatory activity induced by Cramoll 1,4 lectin.…”

“…The ultrastructural analysis confirmed the integrity of follicles cultured in medium supplement with 10 µg/mL PHA. Previous dose response studies have shown that high concentrations of PHA (>80 µg/mL) have toxic effects on cultured fibroblasts, while low concentrations (0.1-10 µg/mL) have no positive effects 30 . In agreement with these data, the present study has demonstrated that the optimal concentration of PHA to stimulate gene expression and antrum formation in cultured preantral follicles is 10 µg/mL.…”

“…According to Nagase et al (2006) this MMP-9 participates in the phenomenon of wound healing, being responsible together with MMP-2 for metabolizing elastin, collagen type IV and other molecules of the ECM. It is known that lectins are chemoattracted by the inflammatory cells and, consequently, stimulate synthesis and secretion of several cytokines involved in the wound healing process (Sell and Costa 2003).…”
