Microplate reader analysis of triatomine saliva effect on erythrocyte aggregation

Abstract: Our hypothesis is that the action of aggregating and disaggregating substances in the blood can be detected and quantified by the Microplate Reader. To ascertain the validity of this hypothesis, we selected two types of blood: one that naturally presents erythrocyte aggregation (pig blood) and the other that does not present aggregation (bovine blood). One important reason for the choice of pig blood is that its erythrocyte aggregation resembles that of human blood. T. infestans saliva was added to the pig blo… Show more

“…Erythrocyte aggregation is characterized by significantly lower absorption values than nonaggregating samples. [35] The positive control caused the formation of aggregates of almost all erythrocytes with absorption (Abs) = 0.051 which was also indicated by the microscopic detection of clusters (stage 3) in Figure 7. In contrast, the negative control (5% glucose containing 10 × 10 -3 m 4-(2-hydroxyethyl)-1-piperazineethanesulfonic acid buffer (GlcHEPES) with erythrocytes) showed the highest Abs value (0.174 ± 0.031) and was classified as stage 1.…”

“…The hematocrit was selected after testing on various hematocrit through different studies (with human, swine and bovine blood samples) using the developed method, the Hct hematocrit = 1,5,10,20,30,40,50 and 60 were tested. In addition, the evaluation of substances such as fibrinogen and albumin [62,63], added in vitro, has also been tested in the past.…”

“…Such structures are known as rouleaux and the process is termed RBC aggregation. Figure 1 shows a representative sample of bovine and porcine RBCs suspended in autologous plasma 1 . Bovine blood does not form rouleaux while porcine blood can form extensive network of rouleaux.…”
