bFGF promotes adipocyte differentiation in human mesenchymal stem cells derived from embryonic stem cells

Abstract: In this work we describe the establishment of mesenchymal stem cells (MSCs) derived from embryonic stem cells (ESCs) and the role of bFGF in adipocyte differentiation. The totipotency of ESCs and MSCs was assessed by immunofluorescence staining and RT-PCR of totipotency factors. MSCs were successfully used to induce osteoblasts, chondrocytes and adipocytes. MSCs that differentiated into adipocytes were stimulated with and without bFGF. The OD/DNA (optical density/content of total DNA) and expression levels of … Show more

“…EGF and bFGF have been shown to improve the adipocyte differentiation capacity of MSCs [43, 45, 46], and bFGF addition during the expansion of hMSCs decreases the expression of the cell marker CD90 [35, 44]. We previously showed that MSCs maintained in serum-free and bFGF/EGF-supplemented medium have low CD90 expression levels [31].…”

“…Further, carrageenan inhibits insulin signalling (Bhattacharyya et al, 2015;Bhattacharyya et al, 2012), which is a potent adipogenic hormone (Klemm et al, 2001). Also, carrageenan may have interacted with FGF-2, which stimulates adipogenic differentiation of mesenchymal stem cells (Kakudo et al, 2007;Kim et al, 2015;Song et al, 2014); after all, a chitosan/ fucoidan hydrogel has been shown to enhance the half-life of FGF-2 (Nakamura et al, 2008) and numerous studies have reported the interaction of www.ecmjournal.org When BMSCs were trypsinised at the end of the pre-condition phase and seeded in new wells for the osteogenic differentiation phase, at day 12, (a) gross visual inspection and (b) DNA quantification revealed that the number of cells that were grown in the presence of MMC in the pre-condition phase (+/− and +/+) was significantly lower then the number of cells that were grown in the absence of MMC in the pre-condition phase (−/− and −/+). (c) The highest calcium content was detected with cells that were grown in the presence of MMC in both pre-condition and differentiation phases (+/+).…”

“…At the pre-implantation stage, murine embryos, human germ cells, and teratocarcinoma stem cells express certain molecular receptors known as Stage Specific Embryonic Antigens (SSEA) on their membrane surface [ 36 ]. SSEA-1 and Nanog occur in embryonic stem cells; both have an important role in maintaining the capacity for self-renewal [ 37 ]. Thus these markers have been used to characterize mouse and human embryonic stem cells [ 38 ].…”
