5S rDNA characterization in twelve Sciaenidae fish species (Teleostei, Perciformes): depicting gene diversity and molecular markers

Abstract: In order to extend the genetic data on the Sciaenidae fish family, the present study had the purpose to characterize PCR-generated 5S rDNA repeats of twelve species of this group through PAGE (Polyacrylamide Gel Electrophoresis) analysis. The results showed the occurrence of at least two different 5S rDNA size classes in all the species. Moreover, 5S rDNA repeats of one of the studied species - Isopisthus parvipinnis - were cloned and subjected to nucleotide sequencing and Southern blot membrane hybridization … Show more

“…To date, the 5S rDNA has scarcely been studied in the Sciaenidae family, and in only one species ( Isopisthus parvipinnis ) two variants of 5S rDNA have been obtained (Alves‐Costa et al . ). Neither of the two NTSs presented homology with the NTS of A. regius , making this region suitable for the designing of species‐specific markers.…”

“…If the whole Sciaenidae family is considered in respect of genetic data, there are only two studies on the cytogenetics of Brazilian species and on 5S rDNA characterization in certain species respectively (Accioly & Molina ; Alves‐Costa et al . ).…”

“…In most eukaryotes, these sequences are organized in tandem repetitions, in which the (NTS)non transcribed spacers present high interspecific variations, due to insertions/deletions, minisatellites and pseudogenes (Nelson and Honda 1985, Leah et al 1990, Alves-Costa et al 2008). However, although evolutionarily conserved, stochastic events may promote a great dispersal of the 5S rDNA sequences in a large number of chromosomes in some Perciformes species (Affonso and Galetti 2005, Lima-Filho et al 2014b).…”

“…The sequences of 5S primers were designed according to Komiya and Takemura [ 53 ]: and ; and those of the 28S primers were: and [ 54 ]. The temperature profile for the amplification of the 5S locus was: initial denaturation step for 5 min at 95°C, followed by 34 cycles (95°C for 15 s, 55°C for 30 s and 72°C for 30 s) and a final extension step at 72°C for 5 min [ 55 ] slightly modified by [ 56 ]. Conditions for PCR of the 28S locus were as follows: initial denaturation step for 3 min at 94°C, followed by 33 cycles (94°C for 30 sec, 53°C for 30 sec and 72°C for 45 sec) with final extension step at 72°C for 10 min [ 57 ] and modified by [ 56 ].…”
