The bone marrow micronucleus test and metronidazole genotoxicity in different strains of mice (Mus musculus)

Abstract: The mouse (Mus musculus) bone marrow micronucleus test was carried out using 24 outbred National Institutes of Health (NIH) mice, 24 inbred Swiss Webster (CFW) mice and 20 inbred Bagg albino/color locus Jackson (BALB/cJ) mice. The mice in the experimental group (n = 32) were injected intraperitoneally with 133 mg kg -1 of metronidazole parenteral solution and the control group consisted of mice (n = 36) which had not been injected with metronidazole. There was no significant difference (p > 0.05) between the s… Show more

“…Mouse femoral bone marrow was obtained from each CF-1 specimens/mice and the micronucleus test was performed as in Schmid (1975), with slight modifications following previous experiences (Abrevaya et al 2007). Mice were killed by cervical dislocation and both femurs extracted.…”

“…The MN index in rodents or human cell is used in the assessment of genotoxicity in vivo or exvivo [5]. In the micro nucleated cells, micronuclei have diameter ≤ 1/3 of intact nucleus and may stained lighter or equal to parent nucleus [6]. The bone marrow cytogenetic study is a short term technique for unrolling the mechanism which is involved in clastogenic and anticlastgenic activities of mutagens [7].…”

“…Micronucleus test is used for studying anti-mutagenic activity of herbal drug [7]. Those cells are considered micro nucleated in which micronuclei are defined chromatin corpuscles with a diameter less or equal to 1/3 of main cell nucleus and stained equally or lighter than the parent nucleus [8]. In micronucleus test using blood erythrocyte, the blood can be collected for extended period of time and chronic and sub-chronic exposure of clastogens can be evaluated for the same animal in the given conditions [9].…”
