Cross-species microsatellite amplification in South American Caimans (Caiman spp and Paleosuchus palpebrosus)

Abstract: Microsatellite DNA markers have been used to assess genetic diversity and to study ecological behavioral characteristics in animals. Although these markers are powerful tools, their development is labor intensive and costly. Thus, before new markers are developed it is important to prospect the use of markers from related species. In the present study we investigated the possibility of using microsatellite markers developed for Alligator mississipiensis and Caiman latirostris in South American crocodilians. Ou… Show more

“…Gels were stained with ethidium bromide and analyzed under UV light. We assumed amplification was successful when we obtained bands with estimated molecular weights within the range established by Zucoloto et al (2006). Products were analyzed by electrophoresis on 10% polyacrylamide gels at 2,200 V and 75 W and stained with AgNO 3 solution (Bassam et al,'91).…”

“…More recently, Villela (2004) recorded high values of genetic diversity and heterozygosity in C. latirostris using microsatellite markers specifically developed for this species Zucoloto, 2003). These broad-snouted caiman primers were also successfully applied to amplification of DNA samples of Paleosuchus palpebrosus, C. crocodylus and C. yacare (Zucoloto et al, 2006). This study aims to utilize microsatellite markers in order to understand the underlying genetic diversity and mating structure of C. latirostris in Santa Fe, Argentina, by following similar work conducted on other crocodilian species.…”

“…Fortunately, a number of microsatellite loci are conserved enough across species to be useful in interspecific and intraspecific marker comparisons (Glenn et al, 1998). Successful cross-species amplification of microsatellites has been reported from groups such as mammals (Williamson et al, 2002;Gunn et al, 2005), birds (Primmer et al, 1996(Primmer et al, , 2005, insects (Wilson et al, 2004), fish (Scribner et al, 1996;King et al, 2001), snakes (King, 2009;Vandewege et al, 2012), and crocodilians (Dever and Densmore III, 2001;Zucoloto et al, 2006;Weaver et al, 2008;Milián-Garcia et al, 2011;Rodriguez et al, 2008Rodriguez et al, , 2011.…”
